Regulation of exosome production and cargo sorting

Abstract: Cellular communication can be mediated by the exchange of biological information, mainly in the form of proteins and RNAs. This can occur when extracellular vesicles, such as exosomes, secreted by a donor cell are internalized by an acceptor cell. Exosomes bear specific repertoires of proteins and RNAs, indicating the existence of mechanisms that control the sorting of molecules into them. Knowledge about loadings and processes and mechanisms of cargo sorting of exosomes is essential to shed light on the physi… Show more

“…Limited cellular specificity has also been observed for some EV regulators evaluated in this study; therefore, it is important to select the most effective activator for each cell type. Furthermore, it has been assumed that there are many pathways by which cargos are sorted onto EVs, and only a few have been uncovered 37 . In this study, EV regulators did not affect the ability of U87MG-EVs or hMSC-EVs to promote angiogenesis or inhibit hypoxia-induced apoptosis.…”

“…Specifically, cargo selectively incorporated into intraluminal vesicles and targeted for secretion via exosomes, is generally recycled from the plasma membrane or delivered from the trans Golgi network [ 46 ]. During exosome biogenesis, cargo sorting can be mediated by several factors, including post-translational modifications of target proteins, such as ubiquitylation [ 47 , 48 ]. These modified proteins can then be recognized by the endosomal sorting complexes required for transport (ESCRTs), in which the ESCRT0 and ESCRTI complexes cluster the ubiquitylated cargo on microdomains of the limiting membrane of multivesicular endosomes [ 46 , 49 ].…”

“…In this way, RNA molecules containing a specific sequence (EXOmotif) are recognized by certain proteins (hnRNPA2B1) that facilitate their entry into exosomes. These proteins also undergo an activation process through reactions known as sumoylation, which helps integration, or isegilation (incorporation of ISG15 into TSG101), which inhibits the generation of exosomes (or facilitates their elimination by fusion to lysosomes) [ 22 , 23 ]. As a result, characteristic miRNAs in exosomes nucleic acid cargoes, while not expressed in the corresponding healthy tissues, have been detected, for instance, in breast cancer [ 24 ] and lung adenocarcinoma [ 25 ], demonstrating their validity as unique disease markers.…”
