Regulation of Follicle-Stimulating Hormone Receptor

Abstract: The acquisition of follicle-stimulating hormone (FSH) receptors during folliculogenesis is believed to be a key event in follicle development. We have examined the effects of FSH and activin on FSH receptor mRNA in cultured rat granulosa cells. Treatment of granulosa cells with FSH resulted in transient suppression of the FSH receptor mRNA levels 2-6 h after treatment, with subsequent recovery at 24 h. We could not detect a similar effect on FSH receptor mRNA by 8-bromoadenosine 3,5-cyclic monophosphate, which… Show more

“…Granulosa cells were washed and collected by brief centrifugation, and cell viability was determined by trypan blue exclusion. The granulosa cells were then cultured in Ham's F-12/DMEM supplemented with 50 mg/l gentamicin sulfate and 0.1% bovine serum albumin on collagen-coated plates in a humidified atmosphere containing 5% CO 2 in 95% air at 37°C [ 19 ].…”

“…We initially used quantitative PCR (qPCR) to examine granulosa cell gene expression in wild-type mice, ␤A flox/Ϫ ;␤B Ϫ/Ϫ mice, and ␤B Ϫ/Ϫ ; ␤A flox/Ϫ ;Amhr2 cre/ϩ mice using a candidate gene approach. FSH receptor (Fshr) and cyclin D2 (Ccnd2) are reported to be upregulated by activin (26)(27)(28)(29). Sequential loss of the activin ␤-subunits reduced Fshr mRNA expression (Fig.…”

“…Rat granulosa cell culture was performed as previously described (Minegishi et al, 1996). Twenty-one-day-old immature Wistar rats were reared in a photoperiod of 14 h of light and 10 h of darkness, with free access to food and water.…”
