Regulation of follicular T helper cell differentiation in antitumor immunity

Abstract: Follicular T helper (Tfh) cells are a subset of CD4+ T cells that play an important role in the formation of germinal centers and the maturation and differentiation of affinity‐matured B cells. Recent studies have demonstrated important functions of Tfh cells in tertiary lymphoid structures of tumors, revealing great potential of Tfh cells in tumor immunity. However, Tfh development is incompletely understood. The differentiation of Tfh cells is a complex, multistage process regulated at the DNA, RNA and prote… Show more

“…Occurrence of CXCL13‐producing Tfh is the sign of TLS maturation. Functional analysis showed that only Th1‐oriented Tfh cells exert adaptive immunity and that this subset is regulated by T follicular regulatory cells (Tfr) 117 . Maintenance of the balance between Tfh and Tfr is a prerequisite for the antitumor effect of TLS 118 .…”

“…Most studies define Tfh cells as CD4 + , CXCR5 + , PD-1 + , ICOS + , BCL6 + or IL-21 + . CXCR5 promotes the migration of Tfh cells into germinal centers [93]. BCL6 is the main transcription regulator in Tfh development [93].…”

“…T follicle helper (Tfh) cells, a subset of CD4 + T cells, are responsible for directing and activating tumor-specific B cells and T cells, thereby promoting the formation and enhancement of antitumor immunity. However, tumors can exploit these cells to suppress the immune response, promoting tumor growth and metastasis (73). METTL14 promotes Th-17 cell differentiation by mediating m 6 A methylation of exosomal miR-149-3p (74).…”
