Regulation of glucose transport as well as glucose transporter and immediate early gene expression in 3T3‐L1 preadipocytes by 8‐bromo‐cAMP

Abstract: In the present study we have examined the ability of 8-bromoadenosine cyclic 3',5'-phosphate (8-bromo-cAMP; the membrane permeant analog of cAMP which can activate protein kinase A) to mimic hormone action and stimulate glucose transport and glucose transporter (GLUT-1) gene expression as well as the expression of several growth-related protooncogenes in quiescent 3T3-L1 fibroblasts. 8-Bromo-cAMP induced a rapid and prolonged increase in the rate of hexose transport. Early activation of hexose transport (withi… Show more

“…A possible explanation for this observation could be that, in addition to the transcriptional control, cAMP regulates hD2 mRNA stabilisation. A similar effect of cAMP on mRNA stability has been demonstrated with several other genes, including GLUT1, GLUT5 and the glucocorticoid receptor (Dong et al 1989, Cornelius et al 1991, Gouyon et al 2003. Interestingly, it has recently been shown (Adams et al 2003) that cAMP prolongs the half-life of human renin mRNA by stimulating the expression of HuR and CP1, two proteins that bind AUUA motifs.…”

“…In the 3T3-L1 fibroblasts, GLUT1 mRNA exhibits a half-life of 42 Ϯ 9 min and behaves much like an immediate-early mRNA (8). However, as the cells differentiate and express the adipose phenotype, a marked stabilization of the half-life occurs increasing to 4.5 Ϯ 1.7 h (21).…”

“…Adipocytes-cAMP has been demonstrated to exert a key role in the control of gene expression in adipocytes (43)(44)(45)(46). Likewise, TNF-␣ has a pleiotropic effect on adipocyte development and metabolism (47,48).…”
