Regulation of LiaRS-Dependent Gene Expression inBacillus subtilis: Identification of Inhibitor Proteins, Regulator Binding Sites, and Target Genes of a Conserved Cell Envelope Stress-Sensing Two-Component System

Abstract: The regulatory network of the cell envelope stress response in Bacillus subtilis involves both extracytoplasmic function -factors and two-component signal transducing systems. One such system, LiaRS, responds to cell wall antibiotics that interfere with the undecaprenol cycle and to perturbation of the cytoplasmic membrane. It is encoded by the last two genes of the liaIHGFSR locus. Here, we analyzed the expression of two LiaRdependent operons, liaIHGFSR and yhcYZ-yhdA, and characterized a palindromic sequence… Show more

“…Our results indicate that SpxB is a novel positive regulator of OatA expression. Interestingly, the spxB promoter region contains a sequence similar to that reportedly recognized by the TCS LiaRS in B. subtilis (38) or CesSR in L. lactis. 10 Both TCSs are members of the envelope stress sensor family and mediate the cellular response to cell wall synthesis-directed antimicrobial treatments in low G ϩ C Gram-positive bacteria (37, 46 -48).…”

“…The homology of SpxB to both TrmA and B. subtilis Spx further suggested its possible regulatory function. By means of MEME motif software (36), we detected a sequence in the spxB promoter region that is similar to a sequence reportedly regulating the expression of the two-component system (TCS) LiaRS in B. subtilis (37,38) or CesSR in L. lactis, 10 both of which respond to cell envelope stress (Fig. 1).…”

“…For example, LiaRS-like systems respond to a variety of cell envelope-damaging agents and usually, but not always, confer resistance to them. 133,141 However, the identity and extent of their target genes varies considerably between species. Another example more pertinent for this review is the ApsXRS/GraXRS system, studied most in staphylococci and streptococci, which responds to CAMPs.…”

“…The cell envelope stress LiaSR two-component regulatory system is widespread among the Firmicutes (Jordan et al, 2006(Jordan et al, , 2008. LiaS is an intramembrane-sensing histidine kinase and believed to sense signals that perturb membrane damage.…”
