Regulation of Membrane Transport by rab GTPases

Abstract: Membrane flow through the cell is a highly dynamic process in which intracellular compartments communicate via tubulo-vesicular structures shuttling cargo molecules to their destinations. Transport carriers are formed at a donor compartment and navigate through the cytoplasm to the target organelle, on which they subsequently dock and fuse. Many of these events are regulated by the cooperative action of monomeric rab GTPases and their effector proteins. Research in recent years resulted in the identification o… Show more

“…The PIM contained 5 g/ml pepstatin, 50 g/ml leupeptin, and 5 g/ml aprotinin and was included with each lysis buffer (31). Samples were analyzed by SDS-PAGE and transferred to nitrocellulose.…”

“…In adipocytes, Rab4 controls recycling of the insulin-regulated glucose transporter GLUT4 (37), and recombinant Rab4 stimulates ␣-granule secretion in platelets in vitro (38). All Rab proteins contain highly conserved domains required for guanine nucleotide binding, GTP/GDP exchange, and GTP hydrolysis that are essential for their proper targeting and function (31).…”

“…In the case of epithelial cells, this includes sorting of proteins to either the apical or basolateral surface of the plasma membrane. The processes by which proteins are sorted into transport vesicles and vesicles are formed, transported, and fused with specific target compartments is mediated by the concerted actions of monomeric Rab proteins, SNARE (soluble n-ethyl-maleimide-sensitive factor attachment protein receptor) proteins and their effectors (Rodman and Wandinger-Ness, 2000;Deneka et al, 2003).…”

“…Rabs, which cycle between GTPbound active states and GDP-bound inactive states as assisted by guanine nucleotide exchange factors or GTPase activating proteins, respectively, are implicated in protein trafficking and targeting and fusion of membranous organelles in all eukaryotic cell types (11,12,40). In acinar cells from the LG, pancreas, and parotid gland, the Rab3D isoform is enriched on mature SV and appears to serve as a negative regulator of homotypic fusion (38).…”
