Regulation of nestin expression by thrombin and cell density in cultures of bone mesenchymal stem cells and radial glial cells

Abstract: Background: Bone marrow stromal cells and radial glia are two stem cell types with neural phenotypic plasticity. Bone marrow mesenchymal stem cells can differentiate into osteocytes, chondrocytes and adipocytes, but can also differentiate into non-mesenchymal cell, i.e. neural cells in appropriate in vivo and in vitro experimental conditions. Likewise, radial glial cells are the progenitors of many neurons in the developing cortex, but can also generate astrocytes. Both cell types express nestin, an intermedia… Show more

“…A similar phenomenon has already been described for glial precursor cells in which serum (through TGFβ factors) inhibited nestin expression [15]. Moreover, we previously demonstrated that thrombin was able to upregulate nestin expression by radial glial cells, but decreased the nestin expression in MSC cell population from bone marrow, indicating that the regulation of nestin expression could be finely modulated in various conditions and/or cell types [12].…”

“…We previously demonstrated that adult rat nestin-positive bone marrow stromal cells isolated from adult bone marrow were able to differentiate into excitable neurons, when co-cultured with mouse cerebellar granule neurons [11]. The proportion of nestin positive-cells in culture was quite low at passage 0, but would increase with reduced cell confluence [12] and higher passage numbers [10], until reaching a plateau (75% nestin- …”

“…Supplementation with FBS (MEM-20%) led to slight expression of a-SMA, whereas the immunoreactivity to nestin is lost. These results are in accordance with previous studies [16][17][18]. Furthermore, and most interestingly, we have demonstrated for the first time that stimulation of hMSC with GBM-conditioned medium results in concentrationdependent expression of desmin, CD 151, VE-cadherin, NG2, and nestin, and significantly increased expression of a-SMA.…”

“…In those conditions, we did observe an increase of the number of nestin-positive cells (presumable NCSC) in early passages. It has been previously shown that nestin expression was dependent on the number of passages [19,20], which allowed nestin-positive cell population to proliferate and take-over. In this study, wnt1 and BMP2 seem to accelerate NCSC proliferation and to favor or to recruit those cells over other cell types, until reaching a plateau after four passages.…”
