Regulation of Phagosome Maturation by Signals from Toll-Like Receptors

Abstract: In higher metazoans, phagocytosis is essential in host defense against microbial pathogens and in clearance of apoptotic cells. Both microbial and apoptotic cells are delivered on a common route from phagosomes to lysosomes for degradation. Here, we found that activation of the Toll-like receptor (TLR) signaling pathway by bacteria, but not apoptotic cells, regulated phagocytosis at multiple steps including internalization and phagosome maturation. Phagocytosis of bacteria was impaired in the absence of TLR si… Show more

“…Apoptotic cells are not trafficked into MHC class II-containing phagosomes 32 , and the inability to properly degrade apoptotic cells has also been linked to autoimmune disease. This opens the possibility that blocking corpse degradation via the Ãstandardà pathway results in inappropriate presentation of antigens derived from apoptotic cells 4 .…”

“…How these candidates coordinately function in phagosome maturation and processing remains to be determined. It is also unknown to what degree proteins identified in these screens would overlap with those containing apoptotic cells, which engage different signaling pathways for internalization and can produce different immunological outcomes 32,33 .…”

“…Impaired phagosome maturation in the absence of MyD88 or TLR4/TLR2 has been shown to impair phagocytosis of bacteria e.g. Escherichia coli, Staphylococcus aureus, and Salmonella typhimurium due to impaired phagosome maturation [6]. Therefore, the bovine mammary epithelial cells possess the capability of dealing with excess bacterial load by up-regulating the necessary mediators of apoptosis.…”

“…Signaling through TLRs has been shown to activate macrophages including enhanced bacterial phagocytosis and phagosome maturation, nucleus translocation of NF-κB and proinflammatory cytokine production [28]. To determine the role of TLR4 in Brucella invasion, uptake of smooth and rough B. abortus was evaluated in BMDMs from C3H/HeJ and C3H/HeOuJ mice.…”
