Regulation of Photosynthesis Genes in Rubrivivax gelatinosus : Transcription Factor PpsR Is Involved in both Negative and Positive Control

Abstract: Induction of biosynthesis of the photosystem in anoxygenic photosynthetic bacteria occurs when the oxygen concentration drops. Control of this induction takes place primarily at the transcriptional level, with photosynthesis genes expressed preferentially under anaerobic conditions. Here, we report analysis of the transcriptional control of two photosynthesis promoters, pucBA and crtI, by the PpsR factor in Rubrivivax gelatinosus. This was accomplished by analyzing the photosystem production in the wild type a… Show more

“…Two potential promoters, P1 and P2, exhibiting an E. coli-like 70 sequence motif were identified at Ϫ126 and Ϫ180 bp from the pucB start codon. Two copies of a conserved repressor PpsR recognition palindrome (TGT-N 12 -ACG/A) at Ϫ137 and Ϫ110 bp from the pucB initiation codon were also identified, involving PpsR in pucBA expression (34). Sequences that might form hairpin structures were found ( Fig.…”

“…This circuit includes the two-component signal transduction system PrrBA (RegBA), the PpsR and FnrL transcriptional factors. So far, in R. gelatinosus only PpsR and FnrL were identified (16,20).…”

“…The roles of PpsR and PpaA have not been clearly defined (9,12,19,34), and biochemical studies of R. denitrificans and other AAPs are needed to clarify their function. Furthermore, AAP genome sequences may reveal patterns in regulatory proteins that could lead to a better understanding of aerobic phototrophy.…”

“…Although R. denitrificans encodes a homolog of PpsR (RD1_0143), aerobic gene repression would be counterproductive to the observed aerobic photosynthetic expression. Interestingly, PpsR in R. gelatinosus and PpsR1 in Bradyrhizobium strain ORS278 have been proposed to activate photosynthesis gene transcription under aerobiosis (19,34).…”
