Regulation of Prostaglandin Synthesis by Interleukin-1β in Cultured Bovine Luteal Cells1

Abstract: Prostaglandins produced within the CL may serve as local modulators of CL function. The present study was designed to characterize the cellular mechanisms by which the cytokine interleukin-1 beta (IL-1 beta) stimulates prostaglandin production in cultured luteal cells. Cycloheximide (CHX) and actinomycin D (Act D) did not affect basal, but completely inhibited IL-1 beta-stimulated prostaglandin F2 alpha (PGF2 alpha) production (p < 0.05). The phospholipase A2 (PLA2) inhibitor, aristolochic acid (PLA2X), and th… Show more

“…In bovine luteal cells (Townson & Pate 1994), IL1B appears to require the stimulation of PL and/or the metabolism of AA for the production of PGs. Moreover, in the bovine uterus, cyclooxygenase-2 (COX2) converts AA to PGH2 (Asselin et al 1997), which is then converted by PGE synthase (PGES) and PGF synthase (PGFS) to PGE2 and PGF2a respectively (Madore et al 2003, Parent & Fortier 2005.…”

“…Third, some components of the intraovarian IL-1 system (e.g., IL-1 ␤ and the type I IL-1 receptor) appear to be expressed in vivo, mainly during a narrow periovulatory window (6)(7)(8)(9)(10)(11)(12). Fourth, IL-1 ␤ has been shown to induce a host of ovulation-associated phenomena in vitro, such as the promotion of prostaglandin production (13)(14)(15)(16), the stimulation of hyaluronic acid biosynthesis (17), the induction of collagenase activity (18), and the activation of nitric oxide synthase activity (19)(20)(21).…”

“…Since IL-1 stimulates ovarian PGE 2 levels (Kokia et al 1992, Brannstrom et al 1993, Townson & Pate 1994, we examined whether this effect was blocked by TGF 1 (Fig. 3).…”

“…It is possible that IL-1 , a promoter of inflammation, and TGF , a promotor of fibrosis and healing, might play complementary roles in the ovulatory process. IL-1 induces a number of inflammatory-like phenomena which have been shown to be associated with ovulation (Kokia et al 1992, Hurwitz et al 1993, Ben-Shlomo et al 1994b, Townson & Pate 1994. In contrast, the function of TGF in ovarian physiology is uncertain (Shull & Doetschman 1994), but evidence suggests that TGF may act as a general regulator of follicle cell differentiation (Dodson & Schomberg 1987).…”
