Regulation of the initiation of DNA replication in human cells

Abstract: The origin of species would not have been possible without high fidelity DNA replication and complex genomes evolved with mechanisms that control the initiation of DNA replication at multiple origins on multiple chromosomes such that the genome is duplicated once and only once. The mechanisms that control the assembly and activation of the replicative helicase and the initiation of DNA replication in yeast and Xenopus egg extract systems have been identified and reviewed [1,2]. The goal of this review is to or… Show more

“…CDC7 kinase and CDKs are the two master S-phase-promoting factors that control origin activation. It is well established that CDKs regulate origin firing with CDK-dependent phosphorylation events regulating origin licensing and usage (reviewed in [57,58]). At the same time, CDKs control the activity of enzymes required for dealing with replication stress, including the CtIP and EXO1 nucleases and the MRN complex [59,60].…”

“…This is in line with recent studies using inhibitors of the upstream kinase ATR, the kinase that activates Chk1. ATR inhibitors synergize with radiotherapy in mouse models by suppressing PD-L1 expression and reducing amounts of T regulatory cells promoting anti-tumor immunity (2). Trials combining ATR inhibitors and PD-1 inhibitors are on the way (e.g.…”

“…Replication is tightly controlled to ensure accurate copying of the large DNA molecule (1). Not only the initiation of replication but also the progression of the replication fork is regulated by kinases (2). If the cell experiences replication stress, UV-induced DNA damage, and enzymatic and helicase remodeling after DNA inter-strand cross-linking chemotherapy or environmental agents, the replication fork is stalled (3).…”
