Regulatory role of miR‐2909 in cell‐mediated immune response

Abstract: Keeping in view the micromanagement of immune response by micro RNAs, the present study was directed to explore the role of miR-2909 in the differentiation and maturation of T-lymphocytes within the population of normal human peripheral blood mononuclear cells maintained in in vitro culture. The results of such a study revealed that miR-2909 had the inherent capacity to significantly increase Treg (CD4+CD25+Foxp3+) cell population and dominant Th1-type cytokine (especially with decrease in IL-4 level and highe… Show more

“…qRT‐PCR results revealed a considerably increased inherent expression of miR‐2909 in both melanoma cells compared to NHEMs (Figure a). This micro‐RNA, besides sustaining its own expression, also ensures the increased expression of UCP2 by downregulating KLF‐4, TGF‐β, and p53, thereby disrupting normal cellular metabolism as evident in our results here (Figure b) and in our previous studies (Kaul et al., ). In the next experimental setup, NHEMs were transfected to ectopically increase the expression of miR‐2909 to understand how it can affect its energy metabolism.…”

“…They either individually or in combination might serve as a prognostic tool to indicate melanoma progression. It is pertinent to note here that apoptosis-antagonizing transcription factor (AATF), in its gene, carries an immunomodulatory micro-RNA, miR-2909, which governs the fate of various genes involved in cellular proliferation, dedifferentiation, and survival as reported in our previous studies (Arora, Kaul, & Sharma, 2014;Kaul, Sasikala, & Raina, 2012). Hence, the present study was addressed to understand whether this novel micro-RNA, miR-2909, has the ability to initiate/elevate/sustain melanoma.…”

“…Micro23 overexpression inhibits Treg generation; in contrast, micro29a overexpression induces Tregs generation. Kaul et al transfected peripheral blood mononuclear cells (PBMCs) with micro2909, which led to an evident increase of CD4 +-CD25 + Foxp3 + Tregs among the CD4 + T population [73]. This indicates that micro2909 may play a regulatory role in Treg proliferation, but this requires further investigation.…”

“…MiRNA-2909 is capable of increasing the cell populations positive for cluster of differentiation 4 (CD4), CD25, and forkhead box P3 (Foxp3), as well as the populations of lymphocytes T helper 1 (Th1). Therefore, this miRNA can help in increasing the number of immune cells capable of protecting against viral infections and other pathogens [ 49 ].…”
