Regulatory Roles for Cytokine- Producing B Cells in Infection and Autoimmune Disease

Abstract: Recent experiments have revealed that B cells can regulate the course of immune responses to pathogens and autoantigens by antibody-independent mechanisms. One antibody-independent function of B cells is to produce cytokines. In this review we describe the identification of IL-10-producing 'regulatory' B cells as well as IFNgamma-producing 'effector' Bel cells and IL-4-producing 'effector' Be2 cells. We discuss the roles of antigen, pathogen-derived molecules and T cell and dendritic cell-derived factors in re… Show more

“…Rather, we show that B cells regulate the emergence of Treg cells in the CNS in a B7-dependent manner. It is becoming increasingly clear that B cells can regulate immune responses in a variety of ways; thus in their absence T cell defects may vary depending on the experimental condition (32,33).…”

“…They are both effective antigen-presenting cells and activators of T cells (33) and also produce proinflammatory cytokines and chemokines (34)(35)(36). Indeed, rituximab treatment in rheumatoid arthritis has been shown to lead to depletion of other major cell populations, such as T cells and macrophages within synovial tissue (37).…”

“…The fact that both surface and intracellular TCR levels are down-regulated in sOVA Tg TCR␣ Ϫ/Ϫ recipients suggests that both internalization and degradation occur. B cells may also inhibit T cell differentiation by producing inhibitory cytokines such as IL-10 and TGF␤ (25,26).…”
