Relationship Between p53 Expression and Other Prognostic Factors in Human Breast Carcinoma:An Immunohistochemical Study

Abstract: Among 843 cases of breast cancer, p53 oncoprotein was detected by the monoclonal antibody (MoAb) Pab-1801 in only 13%. Low-grade carcinomas (tubular, mucinous, papillary, and invasive cribriform types) did not express p53 protein, but it was observed in 4.2% of infiltrating lobular carcinomas (6 of 140 cases) and 50% of pure medullary carcinomas (5 of 10 cases). In intermediate-grade neoplasms, no correlation was seen between p53 status and other putative determinants of a poor prognosis. The latter included h… Show more

“…These results are in keeping with the findings of previous similar studies (Cattoretti et Previous studies have shown that the presence of detectable p53 protein in breast cancer is significantly correlated with increased levels of c-erbB-2 oncoprotein (Iwaya et al, 1991;Barbareschi et al, 1992;Poller et al, 1992;Spandidos et al, 1992;Martinazzi et al, 1993), and EGFR Poller et al, 1992), but similar correlations were not observed in our material. Correlation between Rb and p53 structural abnormalities has been reported in breast carcinoma and in several other malignant tumours (Hall and Lane, 1994).…”

“…Mutation leads to altered protein conformation and increased half-life of the p53 gene product, resulting in detectability by immunohistochemistry. Recent immunohistochemical studies have shown that the detection rate of mutated p53 protein in breast cancer ranges between 13% and 53.5% (Cattoretti et al, 1988;Sawan et al, 1992;Martinazzi et al, 1993;Yamashita et al, 1993).…”

“…The same negative results that were obtained in the current study were shown in 4 of 10 studies concerning lymph node status, 81,90 -92 in 4 of 8 studies concerning patient age, 72,91,93,94 and in 3 of 11 studies concerning tumor size. 58,94,95 The same positive results that were obtained in the current study were found in 2 of 3 studies concerning mitotic activity, 95,96 58,72,77,81,84,87,91,92,95,[97][98][99] and in both studies concerning SPF. 72,95 In the current study, Ki-67 was not correlated with survival, p21 and cyclin A were correlated with survival of any type, and p53 was correlated with MFS.…”

“…The proportion of cancers with p53 mutation (20%), p53 allele loss (41%), p53 mRNA expression (54%) and overexpression (28%) or p53 protein expression (32%) are similar to the reported series (Cattoretti et al, 1988;Davidoff, 1991;Iwaya, 1991;Kovach et al, 1991;Osborne et al, 1991;Runnebaum et al, 1991;Varley et al, 1991;Vojtesek et al, 1992;Andersen et al, 1993;Barnes, 1993;Friedrichs, 1993;Martinazzi, 1993;Thorlacius et al, 1993;Tsuda et al, 1993;Marks et al, 1994;Bergh et al, 1995;Borressen et al, 1995;Stenmark-Askmalm et al, 1995).…”
