Relationship between Platelet PPARs, cAMP Levels, and P-Selectin Expression: Antiplatelet Activity of Natural Products

Abstract: Platelets are no longer considered simply as cells participating in thrombosis. In atherosclerosis, platelets are regulators of multiple processes, with the recruitment of inflammatory cells towards the lesion sites, inflammatory mediators release, and regulation of endothelial function. The antiplatelet therapy has been used for a long time in an effort to prevent and treat cardiovascular diseases. However, limited efficacy in some patients, drug resistance, and side effects are limitations of current antipla… Show more

“…In the literature, it is emphasised that the main components of this cardioprotective diet are tomatoes and their products with antiplatelet function [ 24 27 ]. The bioactivity of tomato extract has been confirmed in both in vitro and ex vivo models [ 10 , 28 33 ]. The results of many experiments show dose-dependent inhibition of platelet aggregation at 8–23% after administration of extract of tomato [ 10 , 29 ].…”

“…However, continuous administration of ASA can induce resistance or side effects, including headache, vomiting, gastric ulceration and abdominal pain in numerous patients (10)(11)(12)(13). Therefore, studies have been investigating naturally derived antiplatelet agents with a reduced risk of side effects (14)(15)(16)(17)(18)(19).…”

“…This has been of special concern in oncology due to the life-threatening consequences of lowered blood levels of drugs, and the potential for severe side effects when blood levels of a drug are increased or actions of herbal products reinforce those of conventional agents. Antiplatelet activity is common in natural products [226], and may aggravate clinical consequences in patients with thrombocytopenia due to chemotherapy or other drugs [227]. Several other examples of negative interactions are known or suspected.…”
