Relationship of Calcium and Membrane Guanylate Cyclase in Adrenocorticotropin-Induced Steroidogenesis*

Abstract: Chlorpromazine, when incubated with isolated adrenal cells, inhibited the ACTH-stimulated formation of cGMP and corticosterone production. It also inhibited the ACTH-stimulated membrane guanylate cyclase, but did not affect the binding of ACTH to the membrane receptors. cGMP-induced steroidogenesis was not affected by the drug. These data indicate that chlorpromazine interferes with adrenal steroid metabolism at a site between the hormone receptor and guanylate cyclase and also show that guanylate cyclase is c… Show more

“…The mechanisms by which hyperammonemia affects steroidogenesis are not clear, but altered function of the glutamate‐nitric oxide cGMP pathway (Hermenegildo et al,1998; Rodrigo et al,2007; Hilgier et al,2009) could contribute to this effect. Steroidogenesis is modulated by nitric oxide (Del Punta et al,1996; Hanke et al,1998; Cymeryng et al,1999) and cGMP (Nambi et al,1982). The HPA axis is also modulated by nitric oxide and cGMP (Kafka et al,1986; Gadek‐Michalska and Bugajski,2008).…”

“…Ca2+-binding proteins, such as calmodulin, play an important role in the secretion mechanism of the adrenal gland (Nambi et al 1982;Widmaier & Hall, 1987;Papadopoulos et al 1990). Kopelman et al (1983) and Gibbons et al (1984) noted the interrela¬ tion of the renin-angiotensin-aldosterone system and the sympathetic nervous system in compensatory response to haemorrhage.…”

“…The reaction was terminated by the addition of 225 μl of 50 mM sodium acetate buffer, pH 6.2, followed by heating on a boiling water bath for 3 min. The amount of cyclic GMP formed was determined by radioimmunoassay (Nambi et al, 1982). All assays were done in triplicate and except where stated otherwise, were performed three times.…”
