Relationship of RNA/DNA ratio and temperature to growth in larvae of Atlantic cod Gadus morhua

Abstract: The RNA, DNA, and protein content of larval cod Gadus morhua reared in the laboratory at 3 temperatures and 3 densities of prey was measured. The data were used to define a quantitative relationship between RNA/DNA ratio (R/D), water temperature (T ), and protein-specific growth rate (SGR, % d -1 ). The nucleic acid content of each larva was determined with both a 2-dye flow-injection fluorometric assay (FIA) and a 1-dye/1-enzyme fluorometric microplate assay (MFA) in order to calibrate each methodology. The r… Show more

“…After standardizing the RD values, data from calibration experiments with Atlantic cod and haddock (Caldarone et al 2003, Caldarone 2005 were reanalyzed and models containing all linear combinations of sRD, T, and sRD × T were constructed (Table 3) Table 3) in relation to fish species using the growth model data set (Table 2). Full species' names in Table 2 0 5 10 15 20 25 30 Temperature ( (Table 2) AIC, the best combined cod and haddock model included the main effect of sRD, the interaction term, and an intercept (Eq.…”

“…In most instances, the addition of terms for larval age, size, or feeding level to these models did not significantly decrease the residual sum of squares, indicating that the relationship among RD, T and G was independent of these variables (e.g. Buckley 1982, Caldarone et al 2003, Caldarone 2005; i.e. while RD and G are known to follow ontogenetic trends (e.g.…”

“…Nucleic acid analysis is an acknowledged practical tool for studying recent overall nutritional condition and growth of larvae and young fish, as well as their responses to environmental variability (e.g. Clemmesen et al, 1997;Chícharo, 1997;Buckley et al, 1999;Caldarone et al, 2003). Specifically, RNA/DNA ratio reflects variations in protein synthesis rates: the quantity of DNA in an animal somatic cell is believed to be normally stable, but the quantity of RNA primarily associated with ribosomes is closely related to the rate of protein synthesis.…”
