Relative recovery of haematopoietic stem cell products after cryogenic storage of up to 19 years

Abstract: There is an increasing trend towards long-term frozen storage of haematopoietic stem cells. For such stem cells, harvested from peripheral blood (PB) or BM, it is not known if stem cell viability decreases with time. In this study, 31 separate bags of stem cell product (SCP) stored for 11-19 years (median 15 years) were assessed for total nucleated cell (TNC) count, colony forming unitgranulocyte/macrophage (CFU-GM), CD34 þ cell count and cell viability. The results were compared with the initial results obtai… Show more

“…Long-term storage (> 60 months) of PBSCs in vapor phase nitrogen after controlled rate freezing also did not have a negative effect on hematopoietic recovery [79]. However, in another study, total nucleated cell count, CD34+ cell count, and cell viability for cells from peripheral blood or bone marrow cryopreserved for 11–19 years was found to decrease with time [82]. …”

“…Besides the relative long-term period of evaluation comparable to other studies [910], our study has three advantages over previous studies assessing CBU quality [910111213]. First, while many previous studies compared groups of CBUs that had been stored for a specific duration, we planned to aliquot a single unit into 21 capillaries that can be tested over time.…”
