RELMα Licenses Macrophages for Damage-Associated Molecular Pattern Activation to Instigate Pulmonary Vascular Remodeling

Abstract: Pulmonary hypertension (PH) is a debilitating disease characterized by remodeling of the lung vasculature. In rodents, resistin-like molecule-a (RELMa, also known as HIMF or FIZZ1) can induce PH, but the signaling mechanisms are still unclear. In this study, we used human lung samples and a hypoxia-induced mouse model of PH. We found that the human homolog of RELMa, human (h) resistin, is upregulated in macrophage-like inflammatory cells from lung tissues of patients with idiopathic PH. Additionally, at PH ons… Show more

“…Another PH-inducing factor, resistin-like molecule-α (RELMα) was increased in a hypoxia-induced mouse model for PH, and its human homologue, resistin, was also found to be upregulated in macrophage-like inflammatory cells in IPAH patients. In these patients, resistin-stimulated macrophages promoted the apoptosis-resistant proliferation of PASMCs [41].…”

“…We then stimulated 3T3-L1 embryonic fibroblasts (ATCC® CL-173™, ATCC, Manassas, VA) with hResistin at different doses for 10 minutes and lysed the cells with Laemmli sample buffer. We analyzed the cell lysates by immunoblotting with anti-phospho-Akt [ 2 , 18 , 20 ] (#4060, Cell Signaling Technology, Danvers, MA) and anti-GAPDH (G8795, Sigma-Aldrich) to determine the activity of the purified hResistin. Western blot analysis was performed as previously described [ 18 ].…”

Western blot analysis was performed as previously described [18]. The Trans-Blot Turbo Nitrocellulose Transfer Kit (#1704271, Bia-Rad) were used and protein bands were visualized by chemiluminescence (ECL; RPN2106, GE Healthcare, Marlborough, MA).…”

“…Furthermore, inflammatory processes are linked to disease progression [ 8 , 16 , 20 ], and plasma levels of cyto- and chemokines such as interleukin (IL)-1β, IL-6, monocyte chemotactic protein (MCP)-1 and tumor necrosis factor (TNF)-α have prognostic value [ 3 , 21 , 39 , 42 ]). A potential source of increased circulating mediators are perivascular infiltrates of mononuclear cells that were described in the lung of PAH patients [ 9 , 29 ]. However, the molecular connection(s) between inflammation and vascular remodeling remain to be deciphered.…”
