Renal Accumulation of Circulating Angiotensin II in Angiotensin II–Infused Rats

Abstract: Previous studies have demonstrated that low-dose angiotensin II (Ang II) infusion for 14 days mimics two-kidney, one clip Goldblatt hypertension and increases intrarenal Ang II levels. The objective of the present study was to determine whether the augmented intrarenal Ang II is due to intrarenal accumulation of the infused Ang II and/or to an increase in intrarenal formation of endogenous Ang II. Male Sprague-Dawley rats were uninephrectomized and divided into three groups: control (N=6), those infused with [… Show more

“…As expected from previous studies, Val5-ANG II elicited the slowly developing hypertension previously seen with 11e5-ANG II. Furthermore, the increases in total intrarenal ANG II were similar to those found in previous studies (62). By using this experimental design, we determined how much of the ANG II came from the minipump and how much of the ANG II was formed endogenously.…”

“…To evaluate mechanisms responsible for the augmented intrarenal ANG II in renin depleted kidneys, further experiments were performed using a model of ANG II infused hypertension that leads to marked suppression of both circulating and tissue renin activity (42,52,62,63). Instead of renal arterial stenosis, an osmotic minipump containing ANG II was implanted.…”

“…[18][19][20][21]24,26,28,29 However, little information is available regarding the prevailing kidney tissue Ang II levels in Cyp1a1-Ren2 rats following induction of either slowly developing or malignant hypertension. In the present study, I3C dosedependently increased PRA and plasma Ang II levels (see Figures 7 and 8).…”

“…In essence, I3C-controlled expression of the Cyp1a1-Ren2 transgene can be considered to be the genetic equivalent of minipump infusion of different fixed amounts of renin that are not subject to homeostatic regulation. Thus, in contrast to the Ang II-infused hypertensive rat model, in which the kidneys of Ang II-infused rats are exposed to elevated circulating Ang II levels but exhibit markedly suppressed plasma renin levels and decreased kidney renin contents, [18][19][20][21] the kidneys of hypertensive Cyp1a1-Ren2 rats are exposed to elevated circulating levels of both renin and Ang II consequent to induction of extrarenal induction of the Cyp1a1-Ren2 transgene. Accordingly, the Cyp1a1-Ren2 transgenic rat can be viewed as a high circulating renin and Ang II hypertensive model in which the elevated blood pressure (BP) is due to increased activity of the circulating or of the extrarenal tissue RAS.…”
