Renal bicarbonate reabsorption in the rat. I. Effects of hypokalemia and carbonic anhydrase.

Abstract: Free-flow micropuncture studies were carried out on superficial rat proximal and distal tubules to assess the participation of different nephron segments in bicarbonate transport. Particular emphasis was placed on the role of the distal tubule, and microcalorimetric methods used to quantitate bicarbonate reabsorption. Experiments were carried out in control conditions, during dietary potassium withdrawal, and after acute intravenous infusions of carbonic anhydrase. We observed highly significant net bicarbonat… Show more

“…In K-depleted animals we have observed in the present study stimulation ofbicarbonate reabsorption in early and late distal tubules, which is compatible with free-flow micropuncture data obtained by Capasso et al (4,5) along the entire distal tubule. This stimulation is inhibited in late distal tubule by SCH 28080, a drug shown to inhibit with great specificity the gastric type K,H-ATPase (45).…”

“…These cells also contain abundant carbonic anhydrase activity (39, 40). Under appropriate conditions, the distal convoluted tubule of the rat has substantial rates of H+ secretion (43). Our results suggest that a proton pump may contribute to H+ transport or to intracellular pH regulation in this segment, but a role for Na+/H+ exchange or other acid/ base transporters has not been excluded.…”

“…Bicarbonate reabsorption along the cortical distal tubule ofthe rat kidney has been studied by both in vivo micropuncture and microperfusion techniques (1)(2)(3)(4)(5). Owing to technical difficulties in isolating these segments, studies in isolated perfused mammalian distal tubules have been scarce (5), and no data are available on bicarbonate reabsorption and acidification.…”
