Renal localization of the constitutive 73-kDa heat-shock protein in normal and PAN rats

Abstract: We purified the constitutive 73-kDa heat-shock protein (HSP73) from the bovine brain, and produced a specific antibody against the protein in a rabbit. On immunoblotting, the antibody cross reacted only with a protein band with a molecular mass of 73 kDa in a crude extract from normal rat kidneys, which was regarded as rat renal HSP73. The intrarenal immunohistochemical distribution of HSP73 was examined by using this antibody, on both normal rat kidneys and kidneys with puromycin aminonucleoside nephrosis. HS… Show more

“…These proteins have been reported to be involved in the folding, assembly, translocation, function, and degradation of intracellular proteins and protein complexes (33)(34)(35). Although high molecular weight hsps (e.g., hsps 60, 72, and 90) have been reported to be present in the kidney (36)(37)(38)(39)(40), localization of hsp27 to the kidney (normal or diseased) has not been previously reported.…”

“…The cells were spun onto slides using Cytospin TM 3 (Shandon Lipshaw Inc., Pittsburgh, PA) and smear specimens of bone marrow were fixed in 100% methanol, dried, and examined by enzymatic immunohistochemistry as previously described [2,7].…”

“…The appearance and the pattern of staining in normal and diseased renal tissues, particularly in distal parts of the nephron, do not represent protein resorption droplets, but probably an enhanced form of constitutive HSP or a combination of constitutive and induced HSP. Perhaps a stronger stimulus (ischemic, hypoxic, or chemi cal) [16,17] may be necessary to produce inducible stress proteins in the proximal portion of the tubules.…”

“…using antibodies against HSP 70 and HSP 85, and in chondrocytes and synovial cells in patients with arthritis [12,13] and in the athero sclerotic plaques of the aorta [ 14] by immunohistochemistry. Similarly, constitutive HSP have been localized in the rat kidneys [15,16]. Dodd et al [17] used antibodies to 72-kD proteins to study human kidney biopsies.…”
