Renin-angiotensin system as a potential target for new therapeutic approaches in Parkinson’s disease

Abstract: Currently, available therapies for Parkinson's disease (PD) are symptomatic. Therefore, the search for neuroprotective drugs remains a top priority. Areas covered: In this review, the potential symptomatic or disease-modifying effect of drugs targeting the Renin-Angiotensin System (RAS) in PD will be explored. Expert opinion: The importance of nigrostriatal local RAS has only begun to be unraveled in the last decades. On one hand, there is a complex feedback cycle between RAS and dopamine (DA). On the other ha… Show more

“…Conversely, longevity was associated with the suppression of this molecule (121). Indeed, ARBs and ACEi were found to be protective against both PD and AD, indicating that RAS may play a key role in neurodegeneration (122)(123)(124). Moreover, in 2017, the US Food and Drug Administration approved a synthetic form of human angiotensin II, Giapreza, for the treatment of septic shock.…”

“…The demonstration of local effects of both AT 2 R and Mas receptor signalling in the brain has led to the exploration of targeting the RAS for many neurological disorders, for example Parkinson's disease [ 216 ], depression [ 217 ], Alzheimer's Disease (AD) [ 218 ], cognitive impairment [ 219 ] or as reviewed here and elsewhere, the cerebrovascular disorder, stroke [ 220 ].…”

“…However, different laboratories have provided solid evidence of the relevant role of RAS in the central nervous system (CNS). Ang is an important regulator of motor control, and AT 1 R and AT 2 R have been suggested as targets to combat Parkinson's disease (PD) and related conditions such as levodopa (L-DOPA)-induced dyskinesias [38,45].…”
