Renin, Cyclooxygenase-2 and Neuronal Nitric Oxide Synthase in the Kidneys of Transgenic Tsukuba Hypertensive Mouse

Abstract: Abstract:The transgenic Tsukuba hypertensive mouse (THM), which expresses the human renin and angiotensinogen genes, develops hypertension secondary to increased renin-angiotensin system activity. The aim of the present study was to assess expression of the renin, cyclooxygenase-2 (COX-2), and neuronal nitric oxide synthase (nNOS) proteins in THM kidneys by immunohistochemical stainings. Renin expression was decreased in the THM kidneys when compared to kidneys from heterozygotes or control mice. Although no d… Show more

“…Parietal layer of the glomerular capsule in male mouse kidney consisted of a single layer of simple cuboidal epithelium, whereas it was single layer of simple squamous epithelium in female. This finding is consistent with the observations of Barberini et al [1] and Yabuki et al [4,3,6]. Vacuoles of different sizes and shapes were observed in the proximal convoluted tubules (PCT) in male, but this histological feature was not detected in female kidney.…”

“…Sex-and strain-dependent differences in the morphology of glomerular capsule have been described in different laboratory animals [5,1,4,7]. DBA/2CrSlc mouse kidney also revealed sex-dependent kidney morphology in the present investigation.…”

“…In the normal human kidney, COX-2 is constitutively expressed in non-MD regions, including glomerular podocytes, small blood vessels, and papillary interstitial cells [13,18]. In contrast, the normal mouse kidney shows minimal expression in MD cells, in the presence or absence of immunopositive signals [10,30]. Although such species-based differences exist between humans and mice, COX-2 expression in MD cells of KK-A y mice closely resembles that of human DN.…”

“…In THM mice, increased expression of COX-2 in the macula densa has been reported [13], and an important role for RAAS in cardiac hypertrophy has been noted [14]. In addition, activation of RAAS has been demonstrated in rats with heart failure [6].…”
