Repeated Administration of Dexamethasone Increases Phosphoinositide‐Specific Phospholipase C Activity and mRNA and Protein Expression of the Phospholipase C β1 Isozyme in Rat Brain

Abstract: Altered hypothalamic-pituitary-adrenal (HPA) function has been shown to be associated with changes in mood and behavior. The enzyme phosphoinositidespecific phospholipase C (PI-PLC), an important component of the PI signal transduction system, plays a major role in mediating various physiological functions. In the present study, we investigated the effects of a single dose and of repeated administration (0.5 or 1.0 mg/kg for 10 days) of dexamethasone (DEX), a synthetic glucocorticoid, on PI-PLC activity and on… Show more

“…Indeed, up to date, only corticoids were reported to regulate the expression of PLC enzymes (Dwivedi & Pandey 1999, Cueille et al 2003. Furthermore, although oestradiol was suggested to facilitate norepinephrine-, CCh-and OTinduced PLC activation at a post-receptor level in the myometrium (Riemer et al 1987, Phaneuf et al 1995, Abdalla et al 2000, no studies have addressed its direct effect on PLC expression.…”

“…The procedures for RNA isolation and competitive RT‐PCR analysis have been described previously (Dwivedi and Pandey 1999). Brain tissues were homogenized in 4 m guanidine isothiocyanate, 50 m m Tris–HCl (pH = 7.4), and 25 m m EDTA, and the total RNA was isolated by CsCl 2 ultracentrifugation.…”

“…PI-PLC activity was measured in both membrane and cytosol fractions by the procedures described earlier by us (Dwivedi and Pandey 1999b) using 5 g protein/ tube in an incubation buffer (20 mM Tris-HCl, 1 mM CaCl 2 , and 100 mM KCl, pH 7.4) containing 10 mM lithium chloride, PIP 2 substrate (50 M unlabeled PIP 2 , 2.0 Ci/ml [ 3 H] PIP 2 , and 0.5 mg/ml certimide) in a total volume of 100 l at 37 Њ C for 10 min. The reaction was terminated by the addition of 500 l of 1 M HCl and 500 l of a mixture of chloroform/methanol (l:l vol/vol).…”
