Replication of respiratory viruses, particularly influenza virus, rhinovirus, and coronavirus in HuH7 hepatocarcinoma cell line

Abstract: Detection of viral antigens and isolation methods has long been used for the diagnosis of respiratory virus infections. The objective was to determine the ability of HuH7 cells to support the replication of prototype and wild strains of respiratory viruses. The cell culture-adapted strains of influenza viruses A and B, parainfluenza viruses 1-4, respiratory syncytial viruses A and B, both strains of the human metapneumoviruses, numerous rhinoviruses, most of the adenoviruses, coronaviruses 229E and OC43, and a… Show more

“…It is a wellestablished human hepatocyte-derived liver cell line model that responds to all types of IFNs and to many cytokines and chemokines (21)(22)(23)(24)(25) and is susceptible to several viruses, including liver and respiratory viruses (7,31). It is important to note that despite the choice of Huh-7 liver cells based on their broad response to a wide variety of IFNs and cytokines, our study is limited by the repertoire of responsive receptors in the liver cells.…”

“…However, our results are consistent with data published by Chang et al (7), which show that Huh-7 cells can harbor a NV RNA replicon, suggesting that these cells are highly susceptible to NV replication. Not only have Huh-7 cells been very successful in allowing replication of other fastidious gastrointestinal viruses such as HCV (29,42,45,46) and hepatitis A virus (28), but they have also been shown to support growth of many respiratory RNA viruses (11).…”
