SUMMARYOchratoxin A (OA), a potent nephrotoxic mycotoxin, was fed to day-old turkey poults at 0 and 4 parts/10 6 dietary levels for 4 and 10 weeks, and its effects, particularly on the immune system, were studied. OA resulted in a significant retardation of growth in both males and females. OA caused a regression of the thymus and bursa of Fabricius and lymphoid depletion in these and other lymphoid organs. Analysis of covariance revealed an enlargement of the kidneys at 10 weeks of age in OA-treated birds; the liver and spleen were enlarged in birds treated with bovine serum albumin (BSA). Cell-mediated immune responses, as measured by delayed hypersensitivity (DH) reactions to avian tuberculin and to BSA in pre-sensitised birds, were significantly depressed in OA-treated birds. The results suggest that the DH immune responses of turkeys differ from those in fowls in some respects; turkeys do not show a typical Arthus reaction and respond poorly to avian tuberculin PPD sensitisation. In a comparative evaluation of skin test sites for the study of DH responses in turkeys, the earskin area was found to be an easy and convenient site for injection, measurement and observation of cutaneous reactions.

Three trials were conducted to assess the effects of adrenocorticotropin (ACTH) and dietary ascorbic acid (AA) on delayed type hypersensitivity (DTH) to human gamma globulin (HGG) in chickens. Broiler chicks received AA at 0, 150, or 300 mg/Kg of feed (ppm) continuously from hatching, were sensitized at 5 wk of age to HGG emulsified in complete Freund's adjuvant, and 2 wk postsensitization, were challenged with an intradermal injection of HGG into a wattle. Birds from each AA group received ACTH at either HGG sensitization, challenge, or both. There were uninjected controls and a vehicle control group, which received gelatin at both sensitization and challenge. The ACTH and gelatin injections were given at 12-h intervals beginning 12 h prior to HGG. Responses to DTH were determined as wattle indices. In all three trials, birds that received ACTH at challenge exhibited a DTH response at 18 and 24 h postchallenge that suppressed, compared with that of controls. Birds that received ACTH at sensitization had a greater wattle response than that of birds that received ACTH at challenge, and this effect was enhanced by dieting AA. In Trial 2, birds that received ACTH at sensitization had a greater wattle response at 18 h post HGG challenge than that of controls. Total leucocyte numbers were unaffected; however, heterophil:lymphocyte ratios were lower in birds that received ACTH at sensitization than in birds that received ACTH at challenge and birds that received ACTH at challenge had fewer lymphocytes. Whether given at challenge or at sensitization, ACTH decreased plasma AA when measured at those times. The 300 ppm level of AA increased plasma AA concentration. Adrenal gland and wattle AA levels were unaffected; however, spleen AA concentration was lower in birds given ACTH at challenge.

Delayed hypersensitivity (DH) reaction to human Ig, corynebacterium parvum or allogeneic cells at the site of tumor cell injection, suppressed the fibrosarcoma (SCFS) growth in chickens. Spleen cells of SC chickens sensitized to human Ig or C. parvum suppressed SCFS growth when adoptively transferred with tumor cells, but only when the sensitizing antigen was present locally. This suppression did not occur in irradiated recipients, SCFS I cells injected into wattles of chickens immune to the tumor, provoked a local DH reaction. Spleen cells from donors sensitized to SCFS I adoptively transferred immunity to both SCFS I and SCFS II when cells of the two tumors were mixed together before injection, but not when SCFS II cells were injected alone. Tumor-specific and nontumor-specific DH may be essential for local suppression of fibrosarcoma growth in chickens.
