Based on the water quality monitoring results from 2002 to 2018 in the coastal waters of Shantou City, this study used eight methods, including the single-factor index method, eutrophication index method, nutrient status quality method, comprehensive index method, organic pollution index method, nitrogen–phosphorus ratio method, potential eutrophication evaluation, and dissolved oxygen saturation parameter method, to evaluate the eutrophication and organic pollution status of the area. The applicability of the first-generation eutrophication model was assessed from the perspective of red tide occurrence, and a comparison was made among the various evaluation methods. The results indicate that the coastal waters of the study area were mostly in a phosphorus-limited state, with a few periods of nutrient deficiency and a very small portion of time in a moderately nutritious state with both nitrogen and phosphorus limitations. The overall N/P ratio showed an increasing trend primarily due to the gradual increase in inorganic nitrogen content. In terms of evaluating eutrophication based on red tide symptoms, the comprehensive index method showed better performance in reflecting eutrophication conditions, while the eutrophication index method and dissolved oxygen saturation parameter method highlighted problematic areas but often exaggerated their extent. The nutrient status quality method and organic pollution index method did not provide distinctive indications and could not verify red tide symptoms. The nitrogen–phosphorus ratio method and potential eutrophication evaluation could reflect certain nutrient structures and eutrophication characteristics but also failed to verify red tide symptoms. Furthermore, this study found a certain correlation between the occurrence of red tide and the ratio of nitrate nitrogen to ammonia nitrogen, which consistently ranged from 1.15 to 1.94 during red tide events. Therefore, whether this ratio can serve as a characteristic value for red tide occurrence and be used for early warning and monitoring still requires further validation.

Tyrosinase is inextricably related to the development of Alzheimer’s disease. The effects of natural tyrosinase inhibitors on human health have attracted widespread attention. This study aimed to isolate and analyze the tyrosinase (TYR) inhibitory peptides in the enzymatic digestion products of royal jelly. We first analyzed optimal process conditions for the enzymatic digestion of royal jelly by single-factor and orthogonal experiments and then used gel filtration chromatography to obtain five fractions (D1~D5) with molecular weights ranging from 600 to 1100 Da. LC-MS/MS was applied to identify the fractions with the highest activity, and the obtained peptides were screened and molecularly docked using AutoDock Vina. The results showed that the optimal enzymatic conditions for tyrosinase inhibition rate were acid protease, enzyme addition 10,000 U/g, initial pH 4, feed-to-liquid ratio 1:4, enzymatic temperature 55 °C, and enzymatic time 4 h. The D4 fraction had the most significant TYR inhibitory activity. The IC50 values of the three new peptides with the strongest TYR inhibitory activity, TIPPPT, IIPFIF, and ILFTLL, were obtained as 7.59 mg/mL, 6.16 mg/mL, and 9.25 mg/mL, respectively. The molecular docking results showed that aromatic and hydrophobic amino acids were more favorable to occupy the catalytic center of TYR. In conclusion, the new peptide extracted from royal jelly has the potential to be used as a natural TYR inhibitory peptide in food products with health-promoting properties.

IntroductionImmunoglobulin G (IgG) is important in mediating humoral immunity and in the maintenance of immune homeostasis in the intestinal mucosa. Oregano essential oil (OEO) is a natural herbal extract that possesses antimicrobial, antioxidant, anti-inflammatory, and immunomodulatory properties. As the effects of OEO on intestinal mucosal immunity in Holstein dairy bulls remained unclear, we investigated the effect of dietary supplementation of OEO on IgG levels and IgG+ cells residing in the intestinal tract in Holstein dairy bulls.MethodsTwelve Holstein bulls in good health of approximately 10 months of age were selected for the experiment and randomly equally divided into two groups. The control (CK) group was fed a basal ration, and in the OEO group, the basal ration was supplemented with OEO (20 g/head/day). After 300 days of feeding, tissue samples of the jejunum, ileum, and colon of the bulls in each group were collected for histopathological analysis, immunohistochemistry, and enzyme-linked immunosorbent assays, respectively.ResultsThe jejunum, ileum, and colon of bulls in the CK group had obvious pathological damage, whereas the structure of each intestinal segment was clear and intact. In the OEO group, pathological damage was significantly reduced. IgG+ plasma cells were diffusely distributed in the lamina propria of the jejunum, ileum, and colon in the CK and OEO groups, with no significant difference between the groups. OEO supplementation significantly reduced the number of IgG+ plasma cells in each intestinal segment, with the highest decrease rate being noted for the ileum (22.87%), followed by the colon (19.45%) and jejunum (8.52%). ELISA test results and immunohistochemical results were mutually verified. The change in IgG content was consistent with the trend of change in the number of IgG+ plasma cells.DiscussionOur findings suggest that OEO supplementation does not alter the diffuse spatial distribution of IgG+ plasma cells in the intestines of Holstein dairy bulls, but lowers immunoglobulin levels to normal levels, significantly reduces intestinal damage, and may enhance mucosal immune defence barrier function by inhibiting inflammatory reactions.
