Background: Snf1, the AMP-activated protein kinase of yeast, is regulated at the level of dephosphorylation. Results: Ligand binding to the Snf1 active site mediates protection from dephosphorylation. Conclusion: Energy charge of the adenosine nucleotide bound at the kinase active site determines rate of dephosphorylation. Significance: Adenylate binding to Snf1 connects kinase activity to energy availability.
