Understanding the genetic architecture of traits in breeding programs can be critical for making genetic progress. Important factors include the number of loci controlling a trait, allele frequencies at those loci, and allele effects in breeding germplasm. To this end, multiparental populations offer many advantages for quantitative trait locus (QTL) analyses compared to biparental populations. These include increased power for QTL detection, the ability to sample a larger number of segregating loci and alleles, and estimation of allele effects across diverse genetic backgrounds. Here, we investigate the genetic architecture of resistance to crown rot disease caused by Phytophthora cactorum in strawberry (Fragaria × ananassa), using connected full-sib families from a breeding population. Clonal replicates of > 1100 seedlings from 139 full-sib families arising from 61 parents were control-inoculated during two consecutive seasons. Subgenome-specific single nucleotide polymorphism (SNP) loci were mapped in allo-octoploid strawberry (2n = 8 × = 56), and FlexQTL software was utilized to perform a Bayesian, pedigree-based QTL analysis. A major locus on linkage group (LG) 7D, which we name FaRPc2, accounts for most of the genetic variation for resistance. Four predominant SNP haplotypes were detected in the FaRPc2 region, two of which are strongly associated with two different levels of resistance, suggesting the presence of multiple resistance alleles. The phenotypic effects of FaRPc2 alleles across trials and across numerous genetic backgrounds make this locus a highly desirable target for genetic improvement of resistance in cultivated strawberry.

The employment of genetic resistance to minimize yield losses induced by Phytophthora capsici remains unexplored in melon (Cucumis melo). A diverse collection of melon accessions was evaluated against P. capsici isolates at the seedling stage. In the first screening assay, 105 accessions were evaluated using isolate PCpe-04 obtained from cucumber (Cucumis sativus). In a second assay, 31 accessions displaying high levels of resistance in the first assay were challenged with a distinct isolate (PCpe-09 also from cucumber). In a third assay, a subset of 14 selected accessions was re-evaluated using isolates PCpe-09 and PCmo-07 (from strawberry). In the last screening, seven accessions with high levels of resistance across all assays were challenged with five isolates from representative host species [PC-Vagem (snap bean), PCp-129 (Capsicum chinense), PCp-155 (C. annuum), PCpe-09 and PCmo-07] to assess their reaction against a varied sample of P. capsici isolates. For two accessions (CNPH-093 and L040), all plants remained free of symptoms after inoculation with all five isolates. Accessions WMR-29, CNPH 084, CNPH 088 and CNPH 092 were also free of symptoms to all isolates, except PCmo-07. These large-spectrum resistance sources might be useful for breeding programs aiming to incorporate resistance against P. capsici in elite melon lines.

Components of genetic variation for resistance of strawberry to Phytophthora cactorum estimated using segregating seedling populations and their parent genotypes

Strawberry ( Fragaria × ananassa ) seedlings from 50 bi-parental crosses among 20 elite genotypes were evaluated for resistance to Phytophthora cactorum after artificial inoculation. Plots of seedlings or runner plants were rated using a disease severity score and the percentage of stunted plants per plot. The distribution of cross means for percentages of plants with stunting was highly skewed; 79% of the inoculated seedlings showed some level of stunting compared to non-inoculated control seedlings, and all but one of the crosses had 50% or more stunted plants. Disease severity scores for the bi-parental crosses were normally distributed and expressed a range of variation not reflected by the percentage of visibly stunted plants. Factorial analysis based on seedling plot means demonstrated significant additive genetic variance for the disease severity score, and the additive genetic variance was 1·9 times greater than the estimated dominance variance. The cross-mean heritability was for the severity score. Estimates of the additive genetic variance component using the covariance of severity scores obtained from the seedling analysis and with severity scores for their parents evaluated in a commercial environment were similar, and 0·30, respectively. Most of the selection response obtained through genotypic selection would thus be transferred to segregating offspring.
