Abstract. Geraldi A, Tay CC, Ni’matuzahroh, Fatimah, Hanafi WNW. 2021. Unraveling the bacterial diversity of Cangar Hot Spring, Indonesia by Next Generation Sequencing of 16S rRNA gene. Biodiversitas 22: 4060-4066. This study is the first attempt at using the Next Generation Sequencing (NGS) method with 16S rRNA to understand the bacterial community structure in an Indonesian hot spring. This study aims to unravel the bacterial diversity of the Cangar Hot Spring as one of the most explored natural hot springs in East Java, Indonesia. We found Proteobacteria and Bacteroidetes as the two most abundant phyla. We discovered the first occurrence of genera Cloacibacterium and Methylobacillus in the hot spring ecosystem, which was the most dominant genera at Cangar Hot Spring. We also found several potential bacteria for bioindustry and bioremediation, such as Acinetobacter junii and Pseudomonas alcaligenes. Besides that, we also observed opportunistic pathogens from genera Comamonas and Vogesella. This study result will provide valuable information for further bioprospecting of bacteria with commercial potential and the development of health and safety measures in the Cangar Hot Spring, among others. Hopefully, this report would encourage the use of NGS technology for studying other hot springs in Indonesia.

Magnetotellurics (MT) is an important geophysical method for exploring geothermal systems, with the Earth resistivity obtained from the MT method proving to be useful for the hydrothermal imaging changes of the system. In this research, we applied the MT method to map the geothermal system of the Seulawah Agam volcano in northern Sumatra, a site intended for the construction of a geothermal power plant with an estimated energy of 230 Mwe. Herein, 3D MT measurements were carried out, covering the entire area of the volcano and the various intersecting local faults from the Seulimeum segment in the NW–SE direction. Based on Occam 2D inversion, a conductive anomaly (<10 ohm·m) near the surface was identified in response to specific manifestation areas, including the Heutsz crater on the northern side and the Cempaga crater on the southern side. A further conductive anomaly was also found at a depth of 1 km, which was presumably due to a clay cap layer covering the fluid in the reservoir layer below the surface, where the manifestation areas are formed at various locations (where faults and fractures are found) owing to the fluid in the reservoir rising to the surface. The MT modeling also revealed that the reservoir layer in Seulawah Agam lies at a depth of 2 km with a higher resistivity of 40–150 ohm·m, which is the main target of geothermal energy exploration. At the same time, the heat source zone where magma is located was estimated to lie in two locations, namely, on the northern side centering on the Heutsz crater area and the southern side in the Cempaga crater area. A clear 3D structure obtained via Occam inversion was also used to visualize the hydrothermal flow in the Seulawah Agam volcano that originates from two heat source zones, where one structure that was consistent across all models is the conductive zone that reaches a depth of 5 km in the south in response to the regional faulting of the Seulimeum segment. Based on the MT research, we concluded that the volcano has the geothermal potential to be tapped into power plant energy in the future.

Magmatic Geothermal Genesis Model in the Huailai Area Based on the Constraints of the Crust–Mantle-Scale Geoelectric Structure

The Huailai area is rich in geothermal resources, but the formation mechanism of its deep heat source is still unclear. In this paper, based on 16 broadband magnetotelluric sounding points, the two-dimensional electrical structure of the crust and mantle in the Huailai area was obtained. Combined with deep seismic reflection and P-wave seismic tomography, the geophysical characteristics of deep heat sources and reservoirs in the Huailai area are described. The Huailai area is characterized by low resistivity and layered reflection above 2 km in depth, which shows the distribution of the Cenozoic sedimentary cover layer. The upper crust is characterized by high resistivity without an obvious reflector, corresponding to the crystalline basement of the basin, whose main lithology is Archean gneiss. There is a highly conductive and bright-spot-reflective structure under the basement, which extends to 100 km, indicating the upwelling of mantle-derived material. Combined with the results of helium isotope tracing, a magma-type geothermal model in the Huailai area is proposed. The upwelling mantle-derived magma material is enriched under the basement to form a heat source. The heat is transferred to the upper crust through heat conduction along the crystalline basement. Then, groundwater circulation brings deep heat to the surface, forming hydrothermal resources.
