Direct interspecies electron transfer (DIET) provides an innovative way to achieve efficient methanogenesis, and this study proposes a new approach to upregulate the DIET pathway by enhancing quorum sensing (QS). Based on longterm reactor performance, QS enhancement achieved more vigorous methanogenesis with 98.7% COD removal efficiency. In the control system, methanogenesis failure occurred at the accumulated acetate of 7420 mg of COD/L and lowered pH of 6.04, and a much lower COD removal of 41.9% was observed. The more significant DIET in QS-enhancing system was supported by higher expression of conductive pili and the c-Cyts cytochrome secretion-related genes, resulting in 12.7-and 10.3-fold improvements. Moreover, QS enhancement also improved the energy production capability, with the increase of F-type and V/A-type ATPase expression by 6.3-and 4.2-fold, and this effect probably provided more energy for nanowires and c-Cyts cytochrome secretion. From the perspective of community structure, QS enhancement increased the abundance of Methanosaeta and Geobacter from 54.3 and 17.6% in the control to 63.0 and 33.8%, respectively. Furthermore, the expression of genes involved in carbon dioxide reduction and alcohol dehydrogenation increased by 0.6-and 7.1fold, respectively. Taken together, this study indicates the positive effects of QS chemicals to stimulate DIET and advances the understanding of the DIET methanogenesis involved in environments such as anaerobic digesters and sediments.

The remediation of polycyclic aromatic hydrocarbon (PAH)-contaminated soils has received much attention in recent years, and most of the contaminated sites are in anaerobic environments, such as deep soils and flooded soils. We simulated the natural flooded soil environment, selected phenanthrene (PHE) as a model PAH contaminant, and designed batch experiments run for 63 days to comprehensively investigate the effects of the combined addition of anaerobic sludge and granular biochar on microbial community and function and the anaerobic biodegradation of PHE. Firstly, the residue, distribution, and removal of PHE in the flooded soil environment were quantified for each group. Secondly, the effects of bioaugmentation of soil indigenous microorganisms by the addition of anaerobic activated sludge and biostimulation of biochar on the removal of PHE from the soil were analyzed against each other. Lastly, the changes in the structure of the microbial community under the effect of bioaugmentation and biostimulation were illustrated by sequencing analyses. The results of this study showed that the removal efficiency of PHE reached 72.0% after the addition of anaerobic activated sludge. The incorporation of anaerobic activated sludge and biochar resulted in a 25.3% increase in PHE removal compared to a single soil, suggesting that the combination of bioaugmentation and biostimulation can have a synergistic effect on the anaerobic biodegradation of PHE in contaminated soils. The results of sequencing analysis further indicated that the introduction of an exogenous microbial community changed the dominant genera associated with PHE degradation and introduced methanogenic archaea, which enriched the metabolic pathways of the carbon cycle in the system. On this basis, the addition of biochar resulted in higher anaerobic microbial community diversity, functional dominant species were enriched, and the direct interspecies electron transfer (DIET) process between electroactive bacteria (Bacteroides, f_Geobacteraceae) and Methanosaeta was facilitated, which accelerated the degradation of PHE by anaerobic microbial communities. The results of this study provide regulatory tools and basic data support for enhanced bioremediation of PAHs in flooded soils.

Residual antibiotics in chicken manure may interfere with the stability of anaerobic digestion (AD) and inhibit resource utilization efficiency. In this study, we aimed to enhance chicken manure bio-methanation in AD with high-concentration of sulfamethazine (SMZ) by adding metal (Fe, Co)-modified Enteromorpha-based hydrochar (Co-HC, Fe-HC). The results showed that Fe-HC and Co-HC increased the degree of acidogenesis by 1.25 times and 1.58 times, respectively. The maximum protein concentration in EPS was increased by 47.64% and 72.5% after adding Fe-HC and Co-HC. However, only Co-HC demonstrated notable improvements in both methane production and SMZ removal efficiency. Electrochemical analysis showed that Co-HC possessed a richer variety of oxygen and nitrogen functional groups, along with superior electron exchange capabilities compared to Fe-HC. Furthermore, microbiological assessments revealed that Co-HC enriched syntrophic bacteria (such as Syntrophomonas and Mesotoga), facilitating direct interspecies electron transfer (DIET) and subsequently enhancing biomethane production. The abundance of genes involved in electron transfer increased significantly with Co-HC, with a maximum increase of 75.86% in Co1.5-HC treatment. Additionally, the elimination of antibiotic resistance genes (sul1, sul2) increased by 65.66% in the Co1.5-HC treatment. This study offers a theoretical foundation and empirical support for the synergistic improvement of livestock and poultry manure containing high antibiotic concentrations, thereby helping to overcome challenges posed by recalcitrant substances.
