Restricted expression of LUZP in neural lineage cells: A study in embryonic stem cells

Abstract: A novel protein LUZP with 3 leucine zipper motifs at its amino terminus is predominantly expressed in the adult brain. A modified gene targeting approach was employed in an attempt to establish in vitro and in vivo models in which Luzp is knock-out (KO) for phenotype assessment and a reporter gene lacZ is knock-in (KI) for tracing its expression. We report in this study the molecular cloning of the Luzp gene, its targeting vector construction and Luzp-KO/lacZ-KI embryonic stem (ES) clone selection. Since LUZP … Show more

“…To determine whether LUZP1 has a role in the establishment of the interaction between ATAC and MED, we purified the ATAC complex from mESCs derived from Luzp1 knockout mice (Lee et al , 2001) and determined the quantity of the MED remaining associated with ATAC (Fig 3B). In Luzp1 −/− mESCs, we observed a marked (∼50%) decrease in the interaction of MED with ATAC (Fig 3C).…”

“…Briefly, genomic DNA containing Gabrg2 was retrieved from the BAC, RP23-233N1, which was generated using DNA isolated from C57BL/6J mice. The targeting vector was prepared by recombineering as described 43 . The targeting vector containing the 5′ long arm, which included exon 8, was approximately 5 kb followed by the LoxP-PGK-neo-LoxP cassette and exon 9 with the Q390X point mutation, which comprised the 3kb 3′ short arm.…”

“…LUZP1 contains an N-terminal LCD1 (Rouse and Jackson, 2000) and three leucine zipper domains ( Fig. 1 A; Sun et al, 1996) and is predominantly expressed in the mouse brain and neural lineages (Lee et al, 2001;Sun et al, 1996). Using antibodies to LUZP1 and centrosome/cilia markers, we found that endogenous LUZP1 localizes to the centrosome, the basal body, actin fibers, and the midbody (Fig.…”
