Restriction fragment length polymorphism of the Vibrio anguillarum serovar O1 virulence plasmid

Abstract: Seventy-eight strains of Vibrio anguillarum serovar 01, all harboring one 65-to 70-kilobase plasmid, were typed according to restriction fragment length polymorphism of the plasmid. Six types, three of which comprised 96% of the strains examined, were produced with the restriction endonuclease BamHI. The fragment length polymorphism type did not correlate to any of 12 different phenotypic properties tested.

“…Comprehensive work has been done to clarify the relatedness of V. anguillarum and V. ordalii Kaper et al 1983;Larsen 1990;Grisez et al 1991;Pazos et al 1993;Alsina et al 1994). These works have shown a clear phenotypic and hence, taxonomic separation between these two species.…”

“…ordalii, as well as comparing these two species. Biochemical characterization (Kaper et al 1983;Larsen 1990;Grisez et al 1991 ;Pazos et al 1993;Alsina et al 1994) and serogrouping (Kitao et al 1983;SBrensen and Larsen 1986;Tajima et al 1986a, b) have probably been the most commonly used methods until now. In the last decade, analysis of bacterial DNA-plasmids as well as chromosomal DNA-has become prominent.…”

“…Following lysis, phenol-chloroform-isoamyl alcohol (25:24:1) was added and the samples were mixed well. After centrifugation, a sample of the supernatant was electrophoresed and plasmids were visualized essentially as described previously (21). V. cholerae O1 V 1057/25 and V 1075/25, each containing an approximately 150-kb plasmid, were used as control strains (27).…”

“…In recent years, restriction fragment length polymorphism (RFLP) of plasmids has become widespread used both in epidemiological studies and the differentiation of several bacterial strains within a species such as Vibrio anguillarum (OLSEN and LARSEN 1990), V. ordalii (PEDERSEN et al 1996) andV. vulnificus (LEWIN et al 2000).…”
