Resveratrol inhibits the hydrogen dioxide-induced apoptosis via Sirt 1 activation in osteoblast cells

Abstract: Sirt 1 plays a critical role in stress responses. We determined the deregulation of Sirt 1 activity, p53 acetylation, Bcl-2 expression, and mitochondria-dependent apoptosis in mouse osteoblast MC3T3-E1 cells which were exposed to H2O2. And then we investigated the protective role of Sirt 1 activator, Resveratrol (RSV), against the H2O2-induced apoptosis. Results demonstrated that Sirt 1 and Bcl-2 were inhibited, whereas p53 acetylation, Bax, and caspase 9 were promoted by H2O2, as was aggravated by the Sirt 1 … Show more

“…Further investigations are needed to explore the relevance of the CRISPR‐cas system and the effect of the E. faecalis strains on apoptosis in osteoblasts. In addition, the expression of Bcl‐2/Bax can be decreased to promote osteoblast apoptosis by glucocorticoids (Lucinda et al., ), sodium fluoride (Yang et al., ), bilirubin (Ruiz‐Gaspa et al., ) and H 2 O 2 (He et al., ), in contrast to the elevated level of Bcl‐2/Bax in saved osteoblast protected by puerarin (Liu et al., ), vaspin (Zhu et al., ) and ghrelin (Liang et al., ). Lipopolysaccharides (LPS) of Escherichia coli can also decrease the Bcl‐2 expression and increase the Bax expression in the MC3T3 cells (Guo et al., ).…”

“…These effects were largely reversed by the addition of Ex-527, a specific inhibitor of Sirt1. The dosage of Ex-527 was selected based on previous reports [20][21][22] and our preliminary experiments.…”

“…In HEK‐293 cells, H 2 O 2 caused a significant reduction in the expression of SIRT 1 and increased expression of SIRT 3. H 2 O 2 has already been shown to inhibit SIRT 1 (He et al, ; Xu et al, ) and increase SIRT 3 (Dai et al, ) expressions. Our study show, for the first time, that SIRT1 is the target of GSPE against H 2 O 2 ‐induced sirtuin dysregulation.…”
