Retinoid X receptor α enhances human cholangiocarcinoma growth through simultaneous activation of Wnt/β‐catenin and nuclear factor‐κB pathways

Abstract: Retinoid X receptor α (RXRα) plays important roles in the malignancy of several cancers such as human prostate tumor, breast cancer, and thyroid tumor. However, its exact functions and molecular mechanisms in cholangiocarcinoma (CCA), a chemoresistant carcinoma with poor prognosis, remain unclear. In this study we found that RXRα was frequently overexpressed in human CCA tissues and CCA cell lines. Downregulation of RXRα led to decreased expression of mitosis‐promoting factors including cyclin D1and cyclin E, … Show more

“…These data are consistent with previous reports indicating the potential tumor suppressor capacity of SOX17 [811]. The Wnt/β-catenin pathway is very relevant in the promotion of cholangiocarcinogenesis [21, 22]. Wnt ligands are highly present in the tumor microenvironment and are released by both cancer-associated fibroblasts (CAFs) and activated macrophages [23].…”

“…This signaling pathway supports the published reports showing that calcium regulates ROS generation (Gorlach et al, 2015). Interestingly, RXRA knockdown was found to decrease cyclin D1 and PCNA expression (Huang et al, 2015), which could also contribute to the induction of cellular senescence.…”

“…IHC was performed as described previously [28]. Briefly, after antigen retrieval using pressure cooker boiling for 2 min, paraffine-embedded human CRC tissue sections were washed and incubated with anti-RARα antibody (1:200) overnight at 4°C followed by incubation with corresponding secondary antibody for 1 h. After washing, the slides were detected with the EliVision Plus Kit (Maixin Bio, Fuzhou, China) according to the manufacturer's instructions and then co-stained with hematoxylin to visualize nuclei.…”
