EDEM2 plays an important role as the first enzyme that acts during mannose trimming of N-glycosylated proteins in the ERAD machinery. Although EDEM2 expression has been reported to be transcriptionally regulated by the IRE1-sXBP1 pathway, very little is known about how endogenous EDEM2 protein expression is regulated. In this work, three different ER stress inducers were used to treat HEK293 cells. Thapsigargan slightly increased both EDEM2 mRNA and protein levels in the cells. Treatment with MG132 did not increase the level of mature EDEM2 protein, and a truncated form of the protein appeared. In SEL1L-deficient cells, there was a slight increase in EDEM2 protein as well as in TXNDC11, a protein that has been reported to form disulfide bonds with EDEM2. On the other hand, EDEM2 protein level decreased in TXNDC11-deficient cells. DTT treatment decreased EDEM2 and TXNDC11 protein levels in a time-dependent manner. The decrease in EDEM2 protein after DTT treatment was attenuated by treatment of the cells with MG132 and by SEL1L deficiency. These findings demonstrate that endogenous EDEM2 protein is regulated posttranscriptionally and that it is in part an SEL1L-mediated ERAD substrate.

Mesencephalic astrocyte‐derived neurotrophic factor (MANF) is a unique member of the neurotrophic factor family residing in the endoplasmic reticulum, where it functions as a stress response protein maintaining endoplasmic reticulum homeostasis, in addition to being secreted extracellularly as a neurotrophic factor to bind with receptors to initiate intracellular signal transduction pathways. Interestingly, MANF has shown an important protective role in the inflammatory response of many diseases. In neural stem cells, pancreatic β cells, and retinal cells, MANF can inhibit the inflammatory response, modulate the immune response, and promote tissue repair. However, the role of MANF in the periodontal inflammatory response remains unclear. In the present study, we used lipopolysaccharide (LPS) from Porphyromonas gingivalis (Pg) to establish a Pg‐LPS‐stimulated periodontal inflammatory model in human gingival fibroblasts cells (HGF‐1) to investigate the role of MANF in vitro. We found that MANF could inhibit pro‐inflammatory cytokine secretion, alleviate the endoplasmic reticulum stress response, promote cell survival, and inhibit cell apoptosis. Therefore, MANF might be a novel promising target for the treatment of periodontitis.

Red blood cell folate level is associated with periodontitis in American adults: results from the NHANES 2009-2014

Background The relationship between folate and periodontal disease has been previously reported. Red blood cell (RBC) folate is an indicator of long-term folate nutrition. The aim of this study was to use the NHANES database to determine whether red blood cell folate is associated with moderate/severe periodontitis. Methods A cross-sectional analysis of 10,168 participants in the NHANES database from 2009–2014 was performed. Multivariate logistic regression was used to determine the OR and 95% CI for periodontitis in each group. Restriction cube spline model was used for analyzing dose-response relationship. Finally, subgroup analysis was performed to test whether the results were robust in different populations. Results After adjusting for confounding factors, there was an inverse correlation between RBC folate concentration and moderate/severe periodontitis. The lowest fraction Q1 (< 360ng/mL) of RBC folate concentration was used as the reference group, multivariable-adjusted ORs and 95% CIs of the second (360-463ng/mL), third (464-569ng/mL), fourth (570–732 ng/mL), and the highest quintile (> 733 ng/mL) categories were 0.87 (0.76–0.99), 0.81(0.70–0.93),0.75(0.65–0.88),0.62(0.53–0.73) respectively (all p < 0.05). Conclusion In representative samples of American adults, high red blood cell folate was at low risk for moderate/severe periodontitis. At the same time, dentists and policymakers should pay attention to the harm caused by high red blood cell folate level.
