Reversal Effect of Dihydromyricetin on Multiple Drug Resistance in SGC7901/5-FU Cells

Abstract: At present, an important therapeutic option for gastric cancer patients is chemotherapy. However, multidrug resistance (MDR) causes the failure of chemotherapy. MDR occurred in cancer cells which are exposed to an anticancer drug and acquired resistance to other multiple anticancer drugs with unrelated structure and mechanism, comparing with the drug-sensitive cancer cells, the intracellular drug concentration in the MDR cancer cells accumulated significantly lowly (

“…The biological properties and the underlying mechanisms of DHM were investigated mostly by in vitro cell cultures and in vivo animal models. In addition, DHM was reported as toxicologically safe and could effectively reverse multidrug resistance [15][16][17][18][19]. Hence, DHM is a promising bioactive compound for developing healthy/functional foods.…”

“…Furthermore, DHM also attenuates the drug efflux mediated by P-gp and then increases the intracellular accumulation of chemotherapeutic drugs, thereby reversing the MDR of human leukemia K562/ADR cells [ 112 ]. In addition, DHM reverses the resistance to 5-FU in SGC7901/5-FU cells in a concentration-dependent manner by downregulating MDR1 mRNA expression and impeding the pumping effect of P-gp [ 113 ], indicating that DHM can be applied as an effective MDR reversal agent in the treatment of gastric cancer. It is known that the survivin gene is a member of the inhibitor of the apoptosis protein family, and the overexpression of survivin may provide growth and survival advantages for cancer initiation and progression [ 114 ].…”

“…Cyclodextrins (CD) are a kind of semi-natural cyclic oligosaccharides with conical hollow structures, which can improve the solubility of drugs. The whole B ring and part of the C ring of DHM could be embedded into the cavity of CD through non-covalent interactions such as hydrogen bonding (YP Wu et al., 2020 ). Therefore, inclusion technology may be an effective strategy to improve the low water solubility of DHM.…”

“…Therefore, the combination with DHM is an effective way to solve the above problems of ADR. In addition to platinum drugs and ADR, DHM can also improve the efficacy of all-trans retinoic acid and reverse the MDR of 5-fluorouracil through various mechanisms (Zhou et al., 2014 ; He et al., 2018 ; M Wu et al., 2020 ).…”
