Rheological Behavior of High Cell Density Pseudomonas putida LS46 Cultures during Production of Medium Chain Length Polyhydroxyalkanoate (PHA) Polymers

Abstract: The rheology of high-cell density (HCD) cultures is an important parameter for its impact on mixing and sparging, process scale-up, and downstream unit operations in bioprocess development. In this work, time-dependent rheological properties of HCD Pseudomonas putida LS46 cultures were monitored for microbial polyhydroxyalkanoate (PHA) production. As the cell density of the fed-batch cultivation increased (0 to 25 g·L−1 cell dry mass, CDM), the apparent viscosity increased nearly nine-fold throughout the fed-b… Show more

“…This trend is in agreement with that observed for P. aeruginosa and Pseudomonas putida . A possible reason for such behavior can be ascribed to shear-induced alignment or breaking of cell aggregates, or eventually to cell deformation as reported in refs and .…”

“…Application of a fed-batch strategy to increase PHA productivity was proved by many other related studies [ 38 , 39 , 40 ]. Therefore, P. resinovorans was cultured with an optimized C/N balance on a flask scale, and cell growth and PHA production monitored ( Figure 6 A).…”

“…The scale-up of agitation speed in IFSBs was validated using the principle of equality of the Froude number. This principle played a pivotal role in propelling the movement of the IFSB, significantly impacting flow characteristics, mixing effects, oxygen transfer, and shear force within the bioreactor (Blunt et al, 2019). Scaling up the total airflow rate in IFSBs was straightforward, as it was based on the working volume.…”
