Ribosomal Proteins and Colorectal Cancer

Abstract: Abstract:The ribosome is essential for protein synthesis. The composition and structure of ribosomes from several organisms have been determined, and it is well documented that ribosomal RNAs (rRNAs) and ribosomal proteins (RPs) constitute this important organelle. Many RPs also fill various roles that are independent of protein biosynthesis, called extraribosomal functions. These functions include DNA replication, transcription and repair, RNA splicing and modification, cell growth and proliferation, regulati… Show more

“…Indeed, RP gene mutations or altered expression have been found in many human tumors (22,23). In this study, based on Ingenuity Pathway Analysis (Ingenuity Systems, www.ingenuity.com), we chose for validation five of them -RPSA, RPS3, RPS7, RPS14, and RPS29 -all involved in cell cycle regulation, apoptosis, and DNA repair.…”

“…It is also an established fact that the phenotypic effects of RP genes extend beyond their canonical ribosomal involvement into extraribosomal functions such as DNA replication, transcription, DNA repair, DNA splicing and modification, and apoptosis [1]. In particular, differential expression of ribosomal proteins (RPs) has also been related to cancers [2, 3]. Recently all ribosomal protein genes have been accorded new nomenclature [4], and this is used in this paper we provide, but the old names are provided at their first mention in the text.…”

“…26 Although different extraribosomal functions have been reported for many r-proteins, but that of RPL18A remains to be elucidated. 27 COII, ND4 and ATP6 are mitochondrial genes involved in oxidative phosphorylation (OXPHOS) biochemical cascade: OXPHOS produces more than 90% of the cellular ATP. The fact that ATP decreases in all cell death mechanisms indicates that energy metabolism might have an important role in tumor cell survival especially under stress conditions including chemotherapy.…”
