Riluzole promotes cell survival and neurite outgrowth in rat sensory neuronesin vitro

Abstract: This study explored the effects of riluzole administration on cell survival and neurite growth in adult and neonatal rat dorsal root ganglion (DRG) neurones in vitro. Neuronal survival was assessed by comparing numbers of remaining neurones in vehicle- and riluzole-treated cultures. A single dose of 0.1 microm riluzole was sufficient to promote neuronal survival in neonatal DRG cultures, whereas repeated riluzole administration was necessary in adult cultures. However, a single administration of riluzole was s… Show more

“…In vitro studies demonstrate that riluzole has the opposite effect of increasing neuronal CGRP expression, together with promoting neurite growth. 47,65 Therefore, it is likely that the reduced CGRP expression in the deep dorsal horn is an indirect consequence of riluzole administration. Aberrant fiber sprouting from the superficial dorsal horn is thought to increase CGRP expression in the deeper laminae.…”

“…Riluzole modulates neuroplasticity via stimulation of growth factor synthesis such as BDNF (Mizuta et al, 2001;Katoh-Semba et al, 2002), promotion of neuritogenesis, neurite branching, and neurite outgrowth (Shortland et al, 2006), increasing concentrations of the neuronal marker N-acetyl-aspartate in hippocampus (Mathew et al, in press), and enhancement of surface expression of the hippocampal AMPA receptor subunits (GluR1 and GluR2) . The latter process is implicated in the regulation of activity-dependent synaptic strength and postsynaptic receptor responsiveness ).…”
