Risks Associated with SGLT2 Inhibitors: An Overview

Abstract: All these drugs are giving promising results in the treatment of diabetes mellitus, but emerging data from post-marketing studies indicate their adverse effects such as diabetic ketoacidosis, genital and urinary tract infection, cancer, bone fracture and foot and leg amputation. Thus, there is a need for better understanding the risk profile of SGLT2 inhibitors. In this review, we have compiled the risk profile of SGLT2 inhibitors by collecting information from various sources such as case reports, published l… Show more

“…Because side effects such as urinary tract infections (UTIs), genital tract infections, and diabetic ketoacidosis (DKA) are predicted on the basis of the mechanism of action of SGLT2 inhibitors, it is necessary to conduct ongoing monitoring of such events [12]. To mitigate their likelihood, a committee of experts has issued recommendations for the appropriate use of SGLT2 inhibitors [13].…”

“…Regarding serious ADRs of special interest, the incidence of cerebrovascular disease in elderly patients was significantly higher than that in non-elderly patients and was consistent with that reported in the STELLA-ELDER study (0.4%). Some previously reported risks of treatment with other SGLT2 inhibitors include amputation, fracture and serious infection of the genital area 30 . In a recent retrospective, single-center, case-control study of T2DM patients, the incidence of lower limb amputations were compared between patients with active diabetic foot wounds who were receiving an SGLT2 inhibitor and those who were not 31 .…”
