RNA interference-mediated control of Asian citrus psyllid, the vector of the huanglongbing bacterial pathogen

Abstract: Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), is the natural vector of the Huanglongbing (HLB) pathogen that presents an unparalleled challenge to citrus production. RNA interference (RNAi) has already shown great potential for insect pest management owing to its high specificity. RNAi technique relies on the delivery of exogenous double-stranded RNAs (dsRNAs) that target essential genes in insects. The D. citri genome possesses three dsRNase homologs and all core RNAi machinery genes,… Show more

“…Thus, the endocytotic mechanism of dsRNA uptake may promote RNAi-mediated gene silencing in organisms. In ACP, the core components of clathrin-mediated endocytosis, namely the clathrin heavy chain and AP50, an AP-2-like protein, have been identified [17].…”

“…For most insects, the results of dsRNA injection delivery differed from those of the other delivery methods. dsRNA injection enables the delivery of a precise amount of dsRNA directly into the target tissues or the hemolymph, consequently achieving a high level of gene silencing [17,56,61]. The first contact before cellular uptake is either the hemolymph or the gut content [31], and these body fluids may act as the main causes of dsRNA instability due to the enzymatic activity of double-stranded ribonucleases (dsRNases) and the physiological pH driving enzymatic activity in these fluids [31,35].…”

“…Research efforts to develop effective management strategies for CLas and CLso have strongly focused on reducing the psyllid population, preventing acquisition and transmission of the bacterial pathogen by using several strategies including: RNAi-based suppression of the psyllid (Hajeri et al, 2014;Taning et al, 2016;Andrade and Hunter, 2017;Ghosh et al, 2017Ghosh et al, , 2018NASEM, 2018;Dubrovina and Kiselev, 2019;Adeyinka et al, 2020;Dubrovina et al, 2020;Yu and Killiny, 2020;Kiselev et al, 2021); antisense oligos, ASO (FANA and Morpholinos) targeting psyllids, their endosymbionts, and bacteria (Wesolowski et al, 2011(Wesolowski et al, , 2013Pietri et al, 2014;Daly et al, 2017;Hunter et al, 2017aHunter et al, ,b, 2018bMetz et al, 2017a,b;Moulton and Moulton, 2017;Patel et al, 2017;Pelz-Stelinski et al, 2017;Hunter and Sinisterra-Hunter, 2018;Xue et al, 2018;Novopashina et al, 2019;Pelz-Stelinski, 2020;Sandoval-Mojica et al, 2020, 2021Hunter and Pelz-Stelinski, 2021); gene-targeting and editing CRISPR/Cas9 systems (Chaverra- Rodriguez et al, 2018;Chaverra Rodriguez et al, 2020;Hunter et al, 2018aHunter et al, , 2019aHunter and Tomich, 2019); insecticides (Tiwari et al, 2011;Grafton-Cardwell et al, 2013;Qureshi et al, 2014;…”

“…The limitations in the availability of efficient, cost-effective strategies for the management of the vector and the disease prompted a large number of investigations for the exploitation of new technologies, and promising results were mainly obtained with RNAi-based approaches (Santos-Ortega & Killiny, 2018;Lu et al, 2019a;Lu et al, 2019b;Yu & Killiny, 2020).…”
