RNA-mediated symmetry breaking enables singular olfactory receptor choice

Ariel D. Pourmorady,
Elizaveta V. Bashkirova,
Andrea M. Chiariello
et al.

Abstract: Olfactory receptor (OR) choice provides an extreme example of allelic competition for transcriptional dominance, where every olfactory neuron stably transcribes one of approximately 2,000 or more OR alleles1,2. OR gene choice is mediated by a multichromosomal enhancer hub that activates transcription at a single OR3,4, followed by OR-translation-dependent feedback that stabilizes this choice5,6. Here, using single-cell genomics, we show formation of many competing hubs with variable enhancer composition, only … Show more

“…In iOSNs, demethylation by LSD1 leads to expression of 5-15 OR genes [67][68][69]. This early coexpression 'primes' the system for the stochastic selection of a single OR later, as one highly expressed OR allele within the group persists and eventually represses competing ORs [56,57,65,[69][70][71][72] (figure 3b).…”

“…Other ORs remain silenced. One OR allele (large green arrow) may be more highly expressed than the others (small green arrows), according to the ‘winner-takes-all’ model [ 56 , 57 ]. ( e ) Left: The Greek Islands form a multi-enhancer hub, which activates the chosen allele through cis and trans interactions.…”

“…It is not known how parasites transition between these transcriptional states; however, this model implies a mechanism that unites the entire var gene family into a single network, potentially providing the basis for coordination of var gene expression within a large population of infected RBCs. Transcriptional dominance is complex and recent evidence in other models, including transcriptional regulation of odorant receptor genes, has identified possible non-coding mechanisms within mRNA that mediate the exclusive expression of a single gene within a large variable gene family [15].…”
