RNAi-Mediated Knockdown of Protein Expression

Abstract: RNA interference is an essential method for studying genomic functions of single genes by loss-of-function experiments. Short interfering siRNAs are efficiently transfected into cultured cells to enable RISC-mediated mRNA cleavage and inhibition of translation in a sequence-specific manner. RNAi enables knockdown of single genes and screening for specific cellular processes or outcomes. In this chapter, we describe a detailed universal protocol for lipoplex-mediated siRNA transfection for cell cultures and cel… Show more

“…It is a post-transcriptional gene regulation mechanism characteristic of (possibly) all eukaryotes, including insect pests [106][107][108]. The mechanism is triggered by double-stranded RNA (dsRNA) precursors that are processed into short-interfering RNA (siRNA) duplexes, which then realize the recognition and repression of complementary dsRNAs, such as mRNAs or viral genomic RNAs [168].…”

“…The transfer of "useful" insectotoxin genes from other economically important Bt strains to endophytic bacteria, as well as the maintenance of their consortiums, should contribute to the creation of new-generation biological agents based on them. At the same time, modern technologies for editing microbial genomes based on the CRISPRCas9 platform [168,175] can be proposed to disable the α-exotoxin and β-exotoxin synthesis of Bt.…”

“…During the last decades, oligonucleotides have become important tools for many applications of biomedical research, and more recently also as therapeutic agents [1]. The modular nature of nucleic acids allows for facile and straightforward, automated synthesis, and their target recognition properties based on hydrogen bonding to the complementary nucleobase confers a number of distinct applications in research and drug development [2][3][4]. Among those, the use as probes for in situ hybridization, in microarrays, for some PCR applications, or the tracking of oligonucleotides in vitro or in vivo requires attachment of fluorescent or other labels to chemically synthesized oligonucleotides [5].…”
