Role of AIB1 for Tamoxifen Resistance in Estrogen Receptor-Positive Breast Cancer Cells

Abstract: Objectives: The p160 nuclear receptor coactivator, AIB1 (amplified in breast cancer 1), is frequently overexpressed in human breast cancer and has been shown to be associated with tamoxifen resistance. The present study aimed to investigate the role of AIB1 in tamoxifen resistance of breast cancer cells. Methods: We reconstructed the RNA interference expression vector, pGenesil-1-U6, specially targeting AIB1 mRNA, and it was stably transfected into the human breast cancer cell line BT474. Cell proliferation an… Show more

“…AIB1 is the only SRC family member that is strongly amplified and overexpressed in human breast cancer and is implicated in tamoxifen resistance (11)(12)(13)(14)(15). Depletion of AIB1 in MCF-7 cells reduces estrogen-mediated cell growth, inhibition of apoptosis, colony formation in soft agar, and tumor growth in nude mice (45).…”

“…The significance of SRCs in breast cancer has been an area of extensive study. AIB1 is the only SRC family member that is strongly amplified and overexpressed in human breast cancer and implicated in tamoxifen resistance (11)(12)(13)(14)(15). AIB1 promotes tumorigenesis and is essential for HER2-driven oncogenesis in mice (16 -18).…”

“…In fact, SRC3 was originally identified as amplified-in-breast cancer 1 (AIB1), which was found overexpressed in breast and ovarian cancers (20). Up-regulation of the SRC3 activity promotes breast cancer growth, invasiveness, and resistance to antiestrogen (21,22). In parallel, SRC3 is found to be overexpressed in prostate cancer cells (23) and is correlated with human prostate cancer seminal vesicle invasion and lymph node metastasis (24).…”
