Role of Dickkopf-1, an Antagonist of the Wnt/β-Catenin Signaling Pathway, in Estrogen-Induced Neuroprotection and Attenuation of Tau Phosphorylation

Abstract: 17␤-Estradiol (E2) has been implicated to be neuroprotective in a variety of neurodegenerative disorders, although the mechanism remains poorly understood. The current study sheds light on this issue by demonstrating that low physiological levels of E2 protects the hippocampus CA1 against global cerebral ischemia by preventing elevation of dickkopf-1 (Dkk1), an antagonist of the Wnt/␤-catenin signaling pathway, which is a principal mediator of neurodegeneration in cerebral ischemia and Alzheimer's disease. E2 … Show more

“…Pla or E2 Alzet minipumps (0.025 mg, 21-d release) were implanted s.c. in the upper midback region under the skin at the time of ovariectomy, and GCI was performed 1 wk later. The dose of E2 used produces serum E2 levels of ∼10-15 pg/mL, which represents physiological low diestrus 1 levels of E2 (37). In some studies, a higher E2 dose was used (0.050 mg, 21-d release), which was administered at the time of ovariectomy (1 wk before ischemia).…”

“…Finally, some LTED animals had E2 replacement initiated immediately and maintained throughout the 10 wk of ovariectomy (10-wk continuous). For GCI, all animals (except sham control) underwent four-vessel occlusion GCI performed as described previously (37). To compare the estrogen receptor levels between intact young and aged Fischer 344 rats, the ovarian cycle was monitored as described previously (13); young cycling rats were used on diestrus 1, and acyclical aged rats showing persistent diestrus were used in the studies.…”

“…Intriguingly, the higher risk and poorer outcome of stroke in postmenopausal women parallels the falling E2 levels that occur after menopause (7,8). Furthermore, exogenous administration of E2 significantly reduces the infarct volume in cortex and hippocampus after focal and global cerebral ischemia (GCI) in various animal models, and female animals exhibit reduced neural damage compared with young adult males after brain injury (1,(9)(10)(11). Taken as a whole, these studies suggest E2 functions as an important neuroprotective factor.…”

“…Crosstalk and synergy between ERα signaling and the WNT pathways have been described (24)(25)(26). In the brain, estrogen signaling activates WNT by down-regulating dickkopf-1 (Dkk1), a WNT antagonist, to prevent neurodegeneration (27). In the uterus, estrogen prompts the canonical WNT signaling pathway in the uterine epithelium to induce uterine epithelial cell growth (28), and in breast cancer, ERα activation enhances cell growth via WNT signaling (29).…”
