Role of Dopamine 2 Receptor in Impaired Drug-Cue Extinction in Adolescent Rats

Abstract: Adolescent drug users display resistance to treatment such as cue exposure therapy (CET), as well as increased liability to relapse. The basis of CET is extinction learning, which involves dopamine signaling in the medial prefrontal cortex (mPFC). This system undergoes dramatic alterations during adolescence. Therefore, we investigated extinction of a cocaine-associated cue in adolescent and adult rats. While cocaine self-administration and lever-alone extinction were not different between the two ages, we obs… Show more

“…They are also implicated in different behaviors. For example, the IL plays a role in inhibition of fear and drug‐seeking (Kim, Li, & Richardson, ; LaLumiere, Niehoff, & Kalivas, ; Sierra‐Mercado, Padilla‐Coreano, & Quirk, ; Zbukvic et al, ), whereas the PL is implicated in the expression of fear and drug‐seeking (Harris, Wimmer, & Aston‐Jones, 2005; Orsini, Kim, Knapska, & Maren, ; Sierra‐Mercado et al, ), at least in males. In females, a dissociation between the PL and IL is also seen, as highlighted by Maeng and Shors () that revealed the PL but not IL is involved in the suppression of learning during a stressful event in female rodents (Maeng & Shors, ).…”

“…Increasingly complex variants of extinction reinstatement have been developed that tease out the importance of contextual (Kim et al ) and discrete drug‐associated cues (Torregrossa et al , ) in driving drug seeking. Indeed, preclinically at least, it is possible to create resistance to relapse by pharmacological manipulation of what is learned during the extinction phase (Perry et al ; Zbukvic et al ). Cognitive aids such as metabotropic glutamate 5 (mGlu5) receptor positive allosteric modulators (PAMs) show potential in this regard (Cleva et al ; Gass & Olive ; Olive ; Perry et al ).…”

“…In fact it is becoming increasingly evident that maturation of the PFC, and in particular changes to dopamine fiber innervation and relative receptor expression, results in divergent mechanisms of cue extinction between adolescents and adults (Zbukvic et al 2016;Zbukvic & Kim, 2018). Therefore it is possible that the discrepancy between current findings in adolescents and previous findings in adults may be mediated through age dependent changes in dopamine system.…”
