The aim of this study was to explore the outcomes of an in ovo GABA injection in broilers challenged with HS. In Experiment 1, 210 Arbor Acres eggs were allocated to five treatments: no-injection, and in ovo injection of 0.6 mL of 0%, 5%, 10%, or 20% of GABA. Hatchling weight and CWEWR were significantly increased in the 5% GABA group. In ovo, injection of 10% GABA solution caused a significant decrease in plasma cholesterol and increased plasma total antioxidant capacity of hatchlings. Experiment 2 was conducted with 126 fertile Arbor Acres eggs distributed into one of two groups. At 17.5 days of incubation, one received no injection, and the other was fed 0.6 mL of 10% GABA. On day 10, one subgroup (4 replicates * 3 birds) from each treatment was submitted to HS (38 ± 1 °C for 3 h) while the other was kept at a thermoneutral temperature (29 ± 1 °C). An in ovo injection of GABA significantly increased total antioxidant capacity, but reduced malondialdehyde levels, hepatic mRNA levels of HSP70, FAS, and L-FABP with HS. In conclusion, an in ovo GABA injection improves CWEWR and antioxidant status at hatch, and enhances antioxidant status while downregulating the expression of HSP70 and fatty acid metabolism-related genes in young chicks under HS.

Revealing the assembly and succession of the chicken gut microbiota is critical for a better understanding of its role in chicken physiology and metabolism. However, few studies have examined dynamic changes of absolute chicken gut microbes using the quantitative microbiome profiling (QMP) method. Here, we revealed the developmental trajectory of the broiler chicken gut bacteriome and mycobiome by combining high‐throughput sequencing with a microbial load quantification assay. We showed that chicken gut microbiota abundance and diversity reached a plateau at 7 days posthatch (DPH), forming segment‐specific community types after 1 DPH. The bacteriome was more impacted by deterministic processes, and the mycobiome was more affected by stochastic processes. We also observed stage‐specific microbes in different gut segments, and three microbial occurrence patterns including “colonization,” “disappearance,” and “core” were defined. The microbial co‐occurrence networks were very different among gut segments, with more positive associations than negative associations. Furthermore, we provided links between the absolute changes in chicken gut microbiota and their serum metabolite variations. Time‐course untargeted metabolomics revealed six metabolite clusters with different changing patterns of abundance. The foregut microbiota had more connections with chicken serum metabolites, and the gut microbes were closely related to chicken lipid and amino acid metabolism. The present study provided a full landscape of chicken gut microbiota development in a quantitative manner, and the associations between gut microbes and chicken serum metabolites further highlight the impact of gut microbiota in chicken growth and development.

The current study explored the influence of dietary supplementation of Chlorella vulgaris dried powder (CV) with zinc-oxide-nanoparticles (ZnO-NPs), and/or selenium-nanoparticles (Se-NPs) on broilers’ growth, antioxidant capacity, immune status, histological responses, and gene expression of some related genes. Several 200 one-day-old Cobb-500 male chicks were distributed into 5 groups with four replicates each. In the 1st group, birds were fed the basal diet (BD). In the 2nd, 3rd, 4th, and 5th groups, birds received the BD supplemented with CV only, CV + ZnO-NPs, CV + Se-NPs, and CV + ZnO-NPs + Se-NPs, respectively. The CV dried powder, ZnO-NPs, and Se-NPs were added to the BD at a rate of 1 g, 40 mg, and 0.3 mg/kg diet, respectively. After 6 weeks of feeding, increases in final body weights (P < 0.05), body weight gain (P < 0.05), and feed intake (P < 0.05) were linked with improvements in FCR (P < 0.05) and intestinal morphometric indices (P < 0.05), and marked up-regulations of MYOS (P < 0.05), GHR (P < 0.05), and IGF (P < 0.05) genes were established. Additionally, distinct increases in antioxidant enzyme activities of SOD (P < 0.05), and GPX (P < 0.05) with increases in the mRNA copies of their genes were measured. Moreover, slight improvement in immunity indices, WBCs count (P > 0.05), and phagocytic and lysozyme activities (P > 0.05) were found. However, distinct increases in phagocytic index (P < 0.05) and up-regulations of IL-1β and TNF, and down-regulation of IL-10 mRNA levels were reported (P < 0.05). These findings were prominent in the case of the separate supplementation of CV with ZnO-NPs or Se-NPs confirming the synergistic mechanisms of CV with ZnO-NPs or Se-NPs. Thus, the synergetic supplementation of CV with ZnO-NPs, or Se-NPs in the broiler’s diet could augment their growth and antioxidant response.
