Role of Macrophages and RhoA Pathway in Atherosclerosis

Abstract: The development, progression, or stabilization of the atherosclerotic plaque depends on the pro-inflammatory and anti-inflammatory macrophages. The influx of the macrophages and the regulation of macrophage phenotype, inflammatory or anti-inflammatory, are controlled by the small GTPase RhoA and its downstream effectors. Therefore, macrophages and the components of the RhoA pathway are attractive targets for anti-atherosclerotic therapies, which would inhibit macrophage influx and inflammatory phenotype, maint… Show more

“…Rho GEFs localize to the cell membrane where they cause activation of the Rho-GTP ( 35 ). Pro-inflammatory and anti-inflammatory macrophages are critical factors for the expansion, progression, or steadying of atherosclerotic plaque ( 5 ). Rho-GTP proteins, especially RhoA, control this process.…”

“…Small GTPases activated by Rho guanine nucleotide exchange factors (GEFs) partly or completely regulate endothelial cell migration and proliferation and adherens junctions of impermeable surfaces. In addition to the endothelium, inflammatory, or anti-inflammatory macrophages controlled by Rho GEFs, the small GTPase RhoA and its downstream effector, and Rho-associated coiled-coil containing kinases (ROCK) are also critical components for the development, regression, or stabilization of the atherosclerotic plaque ( 5 ). The mechanism of atherosclerosis is not fully elucidated.…”

“…Thus, in the CNTN4-RHOA transcript, the first untranslated exon of CNTN4, which is highly expressed in testis, replaced the first untranslated exon of RHOA, bringing the entire coding part of RHOA under control of the CNTN4 promoter (53). RHOA codes for a member of the Rho family of small GTPases with multiple cellular and biological functions (54)(55)(56)(57)(58). Dysregulation of RHOA and other members of the Rho family of small GTPases has been reported in many neoplasias (59,60).…”

“…These LDL modifications are prone to induce immune response; thus, the body forms highly atherogenic circulating LDL, which are involved in immune complexes ( Zhang et al, 2018a ). Other cell types are also involved in AS pathogenesis, such as macrophages and stem cells ( Wang et al, 2015a ; Takamura et al, 2017 ; Ruytinx et al, 2018 ; Kloc et al, 2020 ).…”
